摘  要:CD74,又称恒定链,是一种非多态性II型跨膜糖蛋白,最初认为它是MHC II类分子的分子伴侣,辅助递呈外源性抗原肽,启动体液免疫反应,后来发现CD74作为巨噬细胞迁移抑制因子(MIF)的高亲和力受体,可以介导其生物学效应的发挥,CD74还具有调节淋巴细胞发育及介导胸腺选择等作用。系统性红斑狼疮(SLE)是一种病因不明的累及多系统的自身免疫性疾病,近年来研究发现CD74可能在SLE的发病机制中发挥作用,MIF-CD74信号通路有望成为有效治疗靶点。本文总结了CD74的结构和免疫学功能,以及MIF-CD74信号通路在SLE的发病机制和靶向治疗中的研究进展。CD74,also known as the invariant chain,is a non-polymorphic II type transmembrane glycoprotein. It is a molecular chaperone of MHC II molecules,which assisted in the presentation of exogenous antigen peptides and initiated humoral immune responses. CD74 is also a high-affinity receptor of macrophage migration inhibitory factor(MIF),and mediates its biological effects. In addition,CD7 has a function of regulating lymphocytes development and mediating thymus selection. Systemic lupus erythematosus(SLE)is an autoimmune disease involving multiple systems with unknown etiology. In recent years,studies have found the CD74 plays a role in the pathogenesis of SLE,and the MIF-CD74 signaling pathway is expected to become an effective therapeutic target. This article summarizes the structure and immunological functions of CD74,as well as the progress of research on the MIF-CD74 signaling pathway in the pathogenesis and targeted therapy of SLE.
