摘 要:目的从分子水平研究不同呼吸道上皮细胞感染金黄色葡萄球菌的机制,为临床诊治提供数据依据。方法在GEO基因芯片数据库中得到不同呼吸道上皮细胞感染金黄色葡萄球菌后的表达谱数据,筛选出前50个最显著的差异基因。对差异基因进行GO和Pathway分析,筛选出在不同气道感染情况中金黄色葡萄球菌最显著的信号通路。结果在16HBE14o-和S9细胞株感染表达/非表达Hla毒素金黄色葡萄球菌中,分别筛选1164个和201个显著差异基因(差异大于1.5倍)。差异基因相关的信号通路中,主要是MAPK、JAK/STAT及Toll样受体通路等与免疫反应关系密切。S9细胞株中,主要是FOS蛋白、EGR1蛋白、DUSP1蛋白、ATF3蛋白互作联系紧密;而16HBE14o-细胞株中,蛋白互作不大。结论16HBE14o-和S9细胞株中,表达/非表达Hla毒素金黄色葡萄球菌感染时的基因表达差异显著,S9细胞株的蛋白互作联系紧密。Objective To study the mechanism of different respiratory epithelial cells infected with Staphylococcus aureus at the molecular level to provide data basis for clinical diagnosis and treatment.Methods The expression profile data of different respiratory epithelial cells infected with Staphylococcus aureus were obtained from the GEO gene chip database,and the top 50 most significant difference genes were screened out.GO and Pathway analysis of differential genes were performed to screen out the most significant signal pathways of Staphylococcus aureus in different airway infections.Results In 16HBE14o-and S9 cell lines infected with S.aureus expressing/non-expressing Hla toxin,1164 and 201 significantly different genes were screened respectively(the difference was greater than 1.5 times).Among the signal pathways related to differential genes,the main MAPK,JAK/STAT and Toll-like receptor pathways were closely related to the immune response.In the S9 cell line,the FOS protein,EGR1 protein,DUSP1 protein,and ATF3 protein were interacted closely;while in the 16HBE14o-cell line,the protein interaction was not significant.Conclusion In 16HBE14o-and S9 cell lines,the gene expression difference between expressing and non-expressing Hla toxin S.aureus infection is significant,and the protein interaction of S9 cell line is closely related.
