摘  要:采用四甲基偶氮唑盐(MTT)法检测中国被毛孢提取物及其主要活性成分虫草素对人乳腺癌细胞ZR-75-30的生长抑制作用;采用碘化丙啶(PI)单染荧光检测不同样品对细胞形态及凋亡情况的影响。结果显示,中国被毛袍提取物、虫草素均能抑制人乳腺癌细胞ZR-75-30的生长,IC 50值为中国被毛孢提取物(180.16μg/mL)>虫草素(51.38μg/mL)。通过PI单染荧光拍照检测发现2个样品均以浓度依赖方式诱导人乳腺癌细胞ZR-75-30凋亡,能力大小为虫草素>中国被毛孢提取物。以上结果表明中国被毛孢提取物、虫草素能够有效抑制人乳腺癌细胞ZR-75-30的生长增殖。该研究为中国被毛孢及其活性成分作为癌症辅助治疗手段提供科学依据,为抗肿瘤复方制剂的研究奠定基础。The inhibitory effects of Hirsutella sinensis extract,cordycepin on the cell viability of ZR-75-30 cell were detected by MTT assay.Effect of different samples on cell morphology and apoptosis were observed via PI staining.The experiments showed the Hirsutella sinensis extract and cordycepin had a great inhibitory effect on ZR-75-30 cell in vitro.The IC 50 values of Hirsutella sinensis extract,cordycepin were 180.16 and 51.38μg/mL,respectively.It was found that the two samples induced the apoptosis of ZR-75-30 cell in a concentration dependent manner via PI staining with the order of cordycepin>Hirsutella sinensis extract.The above results suggested that Hirsutella sinensis extract and cordycepin could effectively inhibit the growth and proliferation of ZR-75-30 cell.This study provides a scientific basis for the application of Hirsutella sinensis and its active components as adjuvant treatment of cancer,and lays a foundation for the research of anti-tumor compound preparations.
