摘 要:[目的]建立一种LC-MS/MS测定大鼠血浆中EGCG浓度的方法,进而研究其药动学。[方法]选用Agilent SB-C 18(4.6 mm×250 mm,5μm),乙腈-水(90∶10)为流动相,等度洗脱,ESI离子源,多反应离子监测,负离子扫描;以香草醛为内标,采用内标法定量。[结果]EGCG和内标不受基质干扰;定量限分别为10 ng/mL(EGCG)和5 ng/mL(香草醛),血浆样品批内和批间精密度(RSD)≤11.5%,准确度在92.0%~118.9%,萃取回收率≥92.7%,基质效应为95.3%~115.3%,大鼠腹腔注射20 mg/kg EGCG后的血浆动力学符合二室模型,消除半衰期较长,表观分布容积(Vd)大于总体水体积。[结论]该方法重现性好、分析时间短,能满足EGCG在大鼠血浆中浓度的检测要求,并符合《中国药典》2015年版对生物样品分析的要求,可用于大鼠血浆中EGCG的浓度测定和药代动力学研究。[Objective]To establish a LC-MS/MS method to determine the concentration of EGCG in rat plasma and to study its pharmacokinetics.[Method]Choose Agilent SB-C 18(4.6 mm×250 mm,5μm),acetonitrile-water(90∶10)as mobile phase,isocratic elution,ESI ion source,multi-reactive ion monitoring,negative ion scanning.Vanillin was used as the internal standard,and the internal standard method was used for quantification.[Result]In this method,detection of EGCG and IS were not interfered by the matix,LOQ were 10 ng/mL(EGCG)and 5 ng/mL(vanillin)respectively,the intra-assay and inter-assay precision(RSD)of plasma samples was≤11.5%,the accuracy was 92.0%-118.9%,and the extraction recovery rate≥92.7%,ME was 95.3%-115.3%.After injection of 20 mg/kg EGCG,the plasma dynamics of EGCG was in accordance with the two compartment model,the apparent volume of distribution(Vd)values of EGCG was larger than total body water volume.[Concusion]This method has good reproducibility and short analysis time,can meet the detection requirements of EGCG concentration in rat plasma,and meets the requirements for the analysis of biological samples in the 2015 edition of the Chinese Pharmacopoeia,which can be applied to the pharmacokinetic study of EGCG in rats.
