摘 要:目的:探究博舒替尼对体外人脐静脉血管内皮细胞(HUVECs)缺氧损伤后迁移能力的影响。方法:采用含250μmol/L氯化钴(CoCl_(2))的高糖完全培养基处理HUVECs 24 h用以建立缺氧损伤模型。CCK-8法测定博舒替尼的无细胞毒性浓度。按实验目的分为对照组、CoCl_(2)缺氧损伤组和博舒替尼(0.1μmol/L)治疗组。通过Transwell及划痕实验测定不同组细胞迁移率;蛋白免疫印迹法测定SIK2蛋白含量的变化。使用STRING数据库对SIK2及其相关蛋白做蛋白互作(PPI)分析。结果:0.1μmol/L浓度的博舒替尼可促进缺氧损伤状态下HUVECs的迁移(P<0.05)且降低SIK2的蛋白表达量(P<0.05)。PPI分析结果表明SIK2与血管生成高度相关(P<0.05)。结论:博舒替尼(0.1μmol/L)能提高缺氧状态下血管内皮细胞的存活率、促进细胞迁移能力,并可降低SIK2蛋白表达量。Objective:To explore the effect of bosutinib on the migration ability of human umbilical vein endothelial cells(HUVECs)after hypoxia injury in vitro.Methods:HUVECs were treated with a complete high-sugar medium containing 250μmol/L cobalt chloride(CoCl_(2))for 24 hours to establish a hypoxic injury model.CCK-8 method was used to determine the non-cytotoxic concentration of bosutinib.According to the experiment goal,HUVECs were divided into normal control group,CoCl_(2) hypoxia injury group and bosutinib treatment group.Cell migration rate of different groups was measured by Transwell test and scratch test,and SIK2 protein content was determined by Western blotting.STRING database was used to do PPI analysis of SIK2 related proteins.Results:The 0.1μmol/L bosutinib promoted the migration of HUVECs under hypoxic injury(P<0.05)and reduced the protein expression of SIK2(P<0.05).The results of protein interaction analysis(PPI)indicated that SIK2 was highly correlated with angiogenesis(P<0.05).Conclusion:Bosutinib in dose of 0.1μmol/L can improve the survival rate of vascular endothelial cells under hypoxia,promote cell migration and reduce the expression of SIK2 protein.
