摘 要:旨在研究骨发育相关基因在不同重量鹿茸的尖端表达规律,以期深入研究鹿茸生长发育分子调控机理。利用实时荧光定量PCR技术检测7个基因(MMP14、COL11A1、PHOSPHO1、RHOA、COL1A2、SPARC、CTNNB1)在不同重量鹿茸尖端组织mRNA的相对表达水平,分析骨发育相关基因表达水平与鹿茸重量的相关性;同时利用生物信息软件对目的基因是否存在关联和所参与的生物学过程进行初步研究。结果表明,高重量组鹿茸尖端上述7个基因的mRNA相对表达量都显著高于低重量组,且均与鹿茸重量呈显著正相关(P<0.01或P<0.05)。生物信息分析表明,骨发育相关基因的表达存在互作,MMP14、COL11A1、COL1A2、SPARC、RHOA、CTNNB1共同参与骨骼发育及软骨内骨化。结果提示,高低重量鹿茸的尖端组织中MMP14、COL11A1、COL1A2、SPARC、RHOA、CTNNB1、PHOSPHO1基因的表达存在差异,骨发育相关基因的表达水平会影响鹿茸的重量。The purpose of this experiment was to study the expression of bone development-related genes in the tip part of velvet antlers of different weights so as to further identify the molecular regulation mechanism of velvet antler growth and development.Real-time quantitative PCR was used to detect the relative expression levels of 7 genes(MMP14,COL11 A1,PHOSPHO1,RHOA,COL1 A2,SPARC,CTNNB1)in the tip tissues of velvet antlers of different weights.The correlation between the expression level of the bone development-related genes and the weight of velvet antlers was analyzed.At the same time,bioinformatics software was used to conduct a preliminary study to determine if the target genes had relevance with antler development and if they were involved in the biological processes of the antlers.The results showed that the expression levels of MMP14,COL11 A1,PHOSPHO1,RHOA,COL1 A2,SPARC,and CTNNB1 in the high weight antlers were significantly higher than those in the low weight antlers,and all the expressions were significantly positively related to the weight of antlers(P<0.01 or P<0.05).Bioinformatics analysis showed that there were interactions between the expressions of the bone development-related genes.MMP14,COL11 A1,COL1 A2,SPARC,RHOA,and CTNNB1 all participated in the bone development and cartilage ossification of the antlers.The results indicated that there were differences in the expression levels of the MMP14,COL11 A1,COL1 A2,SPARC,RHOA,CTNNB1,and PHOSPHO1 genes between the tip issues of the high and low yield velvet antlers,and the expression levels of the bone development-related genes had an effect on the weight of velvet antlers.
