摘　　要：目的:建立同时检测大鼠血浆中3种细胞色素P450(CYP450)探针药物,即氯唑沙宗(CYP2E1)、甲苯磺丁脲(CYP2C9)和咪达唑仑(CYP3A4)的高效液相色谱方法。方法:色谱条件选用Topsil TM C18 column(250mm×4.6mm,5μm);流动相为甲醇-(NH4)2HPO4(54∶46,pH值为3.4);检测波长230nm;流速1.0mL·min^(-1);柱温30℃。结果:氯唑沙宗、咪达唑仑、甲苯磺丁脲的线性范围分别为0.2~50,0.2~50,0.1~50μg·mL^(-1);方法回收率分别为96.22%~101.42%,98.72%~104.14%,97.09%~105.27%。结论:该HPLC检测方法快捷,准确,灵敏度高,符合生物样品分析要求,适用于同时测定3种CYP450酶亚型探针药物的大鼠血浆药物浓度,可为相关肝药酶代谢研究提供方法学参考。OBJECT To establish a method for simultaneously determining three CYP450s probe drugs,chlorzoxazone(CYP2E1),tolbutamide(CYP2C9)and midazolam(CYP3A4),in single run in rat plasma.METHOD The isolation was performed on a Topsil TM C18 column(250mm×4.6mm,5μm)with mobile phase consisting of methanol-(NH4)2HPO4 buffer(54∶46;pH 3.4)at the flow rate was 1.0mL·min^(-1).The detection wavelength was 230nm;and the column temperature was 30℃.RESULT There were good linearity of chlorzoxazone,tolbutamide and midazolam in the ranges of 0.2~50,0.2~50,0.1~50μg·mL^(-1).The recoveries were 96.22%~101.42%,98.72%~104.14%,97.09%~105.27%,respectively.CONCLUSION The method is reliable,sensitive and suitable for simultaneously determining the concentrations of three CYP450 probe drugs in rat plasma,which could be employed in evaluating the effects of drugs on CYP450 subtype enzymes activities and in relatived studies.
