摘 要:目的优化楮实子多糖提取工艺,考察其抗皮肤光老化能力。方法以多糖得率为指标,采用水提醇沉法提取楮实子多糖,在单因素基础上利用L9(34)正交试验优选楮实子多糖的提取工艺。通过人皮肤成纤维细胞建立的光老化模型考察楮实子多糖抗光老化作用。结果最终确定的最佳提取工艺条件为:提取温度80℃,提取次数2次,料液比为1∶20(g∶mL)。楮实子多糖能够显著增加超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)水平,降低丙二醛(MDA)水平。结论楮实子多糖的提取工艺合理可行,楮实子多糖有抗UVB致皮肤成纤维细胞氧化损伤作用,可为其深入研究提供参考。OBJECTIVE To optimize the extraction technology of polysaccharides from Fructus Broussonetiae and investigate its anti skin photoaging activity.METHODS The extraction process of Polysaccharides from Fructus Broussonetiae by water extraction and alcohol precipitation method.Based on the single factor and L9(34)orthogonal test,the extraction process of Fructus Broussonetiae polysaccharides was optimized with the yield of polysaccharide as the index.The anti photoaging ability of Fructus Broussonetiae polysaccharides was investigated by establishing a photoaging model of human skin fibroblasts.RESULTS The optimal extraction conditions were determined as follows:the extraction temperature of 80℃,the extraction times of twice,the solid-liquid ratio of 1∶20(g∶mL).The Fructus Broussonetiae polysaccharides could significantly increase the levels of superoxide dismutase(SOD)and Glutathione peroxidase(GSH-PX),and reduce the level of malondialdehyde(MDA).CONCLUSION The optimized extraction technology of polysaccharides from Fructus Broussonetiae is reasonable and feasible.Fructus Broussonetiae polysaccharides have anti UVB induced oxidative damage of skin fibroblasts and can provide reference for further study.
