摘 要:目的利用复发性流产(RM)小鼠模型探讨联合使用孕鼠中分离的调节性T(Treg)细胞和维生素D_(3)治疗RM的效果。方法将RM小鼠分为6组(联合治疗组、Treg细胞组、维生素D_(3)组、生理盐水组、空白对照组及正常妊娠组),分别行不同方式治疗,观察6组间外周血Treg和Th17细胞比率、Treg细胞联合维生素D_(3)治疗对相关基因表达情况及胚胎吸收率。结果联合治疗组与阴性对照相比在Treg、Th17细胞及两者的比值均具有显著差异,与单独Treg细胞或维生素D_(3)组相比三项参数也有显著差异(P<0.05);Q-PCR和Western Blot分析表明联合治疗组Foxp3表达量最多,单独Treg细胞或维生素D_(3)组表达水平次之,阴性对照最少,VDR的表达情况与Foxp3类似,单独维生素D_(3)组与联合治疗组表达量无差异(P>0.05),联合治疗组、单独Treg或维生素D_(3)的CYP27B1的表达与阴性对照相比有显著差异(P<0.05);在胚胎吸收率上,联合治疗组与单独维生素D_(3)、阴性对照相比有显著差异(P<0.05)。结论对于RM小鼠给予孕前注射Treg细胞和孕中补充维生素D_(3)的联合治疗方式的效果优于单独补充的方式,可成为免疫异常的RM患者治疗的新选择。Objective To investigate the effect of combined use of regulatory T(Treg)cells isolated from pregnant mice and vitamin D3 in the treatment of recurrent spontaneous abortion(RM).Methods RM mice were divided into 6 groups:the combined treatment group,the Treg cell group,the vitamin D3 group,the normal saline group,the blank control group and the normal pregnancy group,and treated with different methods respectively.The ratio of Treg and Th17 cells in peripheral blood,the expression of related genes and embryo absorption rate of Treg cells combined with vitamin D3 treatment were observed.Results There were significant differences in Treg,Th17 cells and their ratio between the combined treatment group and the negative control group,and there were also significant differences in the three parameters compared with Treg cells alone or vitamin D3 group(P<0.05);Q-PCR and Western blot analysis showed that the expression of Foxp3 was the highest in the combined treatment group,followed by Treg cells alone or vitamin D3 group,the lowest in the negative control group,and the expression of VDR was similar to that of Foxp3,there was no difference in the expression of CYP27B1 between the vitamin D3 group and the combined treatment group(P>0.05),but there was significant difference in the expression of CYP27B1 between the combined treatment group,Treg or vitamin D3 group and the negative control group(P<0.05),and there was significant difference in the embryo absorption rate between the combined treatment group and the vitamin D3 group and the negative control group(P<0.05).Conclusion For RM mice,the effect of Treg cells injection before pregnancy and vitamin D3 supplement during pregnancy is better than that of single supplement,which can be a new choice for RM patients with immune abnormalities.
