摘  要:本试验旨在研究葡萄籽原花青素提取物(GSPE)对大鼠脂肪沉积及脂质代谢的影响。试验选取雌性SD大鼠52只(170~220 g),适应性饲养1周后将其随机分为对照组(n=22)和高脂组(n=30)。通过向高脂组大鼠饲喂高脂饲粮建立大鼠肥胖模型,8周后高脂组肥胖模型建立成功,淘汰肥胖抵抗型大鼠后将对照组随机分为Ⅰ组、Ⅱ组,高脂组随机分为Ⅲ组、Ⅳ组,每组10个重复,1个重复1只。将GSPE用生理盐水溶解,Ⅰ组、Ⅲ组大鼠用生理盐水灌胃,Ⅱ组、Ⅳ组大鼠均按500 mg/kg体重剂量用GSPE溶液灌胃。试验期5周。结果表明:1)高脂饲粮与GSPE对肝脏重量的影响存在显著交互作用(P<0.05),对白色脂肪组织(WAT)重量的影响存在交互作用趋势(0.05<P<0.10);GSPE能够显著降低肥胖大鼠的肝脏重量及WAT重量(P<0.05),对脾脏、肾脏重量无显著影响(P>0.05)。2)高脂饲粮与GSPE对血清高密度脂蛋白胆固醇(HDL⁃C)及低密度脂蛋白胆固醇(LDL⁃C)含量的影响存在显著交互作用(P<0.05),GSPE能够显著提高肥胖大鼠血清中HDL⁃C含量并显著降低LDL⁃C含量(P<0.05)。3)高脂饲粮与GSPE对WAT中过氧化物酶体增殖剂激活受体γ(PPARγ)基因相对表达量的影响存在显著交互作用(P<0.05),对脂肪酸合成酶(FAS)基因相对表达量的影响存在交互作用趋势(0.05<P<0.10),GSPE能够显著下调肥胖大鼠WAT中脂肪合成基因PPARγ和FAS基因相对表达量(P<0.05)。4)高脂饲粮与GSPE对WAT中无翅型MMTV整合位点家族3a(Wnt3a)及β-连环蛋白(β⁃catenin)含量的影响均存在显著交互作用(P<0.05),GSPE能够显著下调肥胖大鼠WAT中Wnt3a及β⁃catenin的含量(P<0.05)。由此可见,GSPE具有促进肥胖模型SD大鼠脂质代谢并抑制其脂肪沉积作用;GSPE的降脂作用可能与Wnt3a/β⁃catenin信号通路有关。The aim of the present study was to evaluate the effects of grape seed proanthocyanidin extract(GSPE)on fat deposition and lipid metabolism in obese SD rats.A total of 52 female SD rats(170 to 220 g)were selected for the experiment,and they were randomly divided into control group(n=22)and high⁃fat group(n=30)after 1 week of adaptive feeding.A rat obesity model was established by feeding high⁃fat diets to the rats of high⁃fat group.After 8 weeks,the obesity model of the high⁃fat group was successfully estab⁃lished.After the obesity⁃resistant rats were eliminated,the control group was randomly divided into groupsⅠandⅡ,and the high⁃fat group was randomly divided into groupsⅢandⅣ,each group had 10 repetitions and 1 rat for each repeat.The GSPE was dissolved in normal saline,rats in groupsⅠandⅢwere gavaged with normal saline,rats in groupsⅡandⅣwere gavaged with GSPE solution,and each group was gavaged at a dose of 500 mg/kg BW GSPE.The trial period was 5 weeks.The results showed as follows:1)there was a significant interaction effect between the high⁃fat diet and the GSPE on liver weight(P<0.05),and there was a trend of interaction effect on the influence of white adipose tissue(WAT)weight(0.05<P<0.10).GSPE significantly reduced the body weight,liver weight and WAT weight of obese rats(P<0.05),and had no sig⁃nificant effect on the weight of spleen and kidney(P>0.05).2)There was a significant interaction effect be⁃tween the high⁃fat diet and the GSPE on high density lipoprotein cholesterol(HDL⁃C)and low density lipopro⁃tein cholesterol(LDL⁃C)contents in serum(P<0.05).GSPE significantly increased the HDL⁃C content in the serum of obese rats and significantly decreased the LDL⁃C content(P<0.05).3)There was a significant inter⁃action effect between the high⁃fat diet and the GSPE on the relative expression level of peroxisome proliferator activates receptorγ(PPARγ)mRNA in WAT(P<0.05),and there was a trend of interaction effect on the relative expression level of fatty
