摘　　要：试验研究蒙兽药巴布-7成分复方(BSEIC)对腹泻小鼠肠道结构及屏障功能相关因素的影响。选取60只40日龄昆明系SPF小鼠随机分为6组,即NC组(生理盐水)、UC组(2%Na HCO3+E.coli O8)、CIP组(2%Na HCO3+E.coli O8+环丙沙星0.50 mg/kg BW)、BSEIC.H组(2%Na HCO3+E.coli O8+BSEIC 1 g/kg BW)、BSEIC.M组(2%Na HCO3+E.coli O8+BSEIC 0.5 g/kg BW)、BSEIC.L组(2%Na HCO3+E.coli O8+BSEIC0.25 g/kg BW)。2%Na HCO3+E.coli O8造腹泻模型3 d,药物治疗3 d,统计计算造模后和治疗后稀便率及稀便指数。治疗结束后禁食24 h处死小鼠,采集血清及回肠,ELISA法检测血清中肿瘤坏死因子-α(TNF-α)、白介素-4 (IL-4)、白介素-6 (IL-6)和白介素-10 (IL-10)含量及回肠中分泌型免疫球蛋白A (SIg A)含量。HE染色法观察回肠组织结构变化,PAS染色观察回肠中杯状细胞形态及数量的变化。结果显示,BSEIC高、中、低剂量均能降低小鼠腹泻率与腹泻指数,恢复腹泻模型小鼠回肠形态,提高回肠杯状细胞数量,降低血液中IL-6和TNF-α含量,提高IL-4、IL-10以及s Ig A含量。试验表明,蒙兽药BSEIC复方可以改善E.coli O8所致小鼠肠道屏障损伤。The experiment studied the effect of ingredients compound of mongolian veterinary medicine Babu-7(BSEIC) on intestinal structure and barrier function of diarrhea mice.60 40-day-old Kunming SPF mice were randomly divided into six groups,namely NC group (normal saline),UC group (2%Na HCO3+E.coli O8),CIP group (2%Na HCO3+E.coli O8+ciproxazole Star 0.50 mg/kg BW),BSEIC.H group (2%Na HCO3+E.coli O8+BSEIC 1 g/kg BW),BSEIC.Mgroup (2%Na HCO3+E.coli O8+BSEIC 0.5 g/kg BW),BSEIC.L group (2%Na HCO3+E.coli O8+BSEIC 0.25 g/kg BW).2%Na HCO3+E.coli O8was used to create a diarrhea model for 3 d,followed by medical treatment for 3 d,and statistically calculate the loose stool rate and loose stool index after modeling and treatment.After the treatment,the mice were sacrificed by fasting for 24 h,and the serum and ileum were collected.The serum TNF-α,IL-4,IL-6 and IL-10 content and the ileum SIg A content were detected by ELISA method.The ileum tissue structure was observed by HE staining method Change.PAS staining was used to observe the changes in the morphology and number of goblet cells in the ileum.The results showed that high,medium and low doses of BSEIC can reduce the rate of diarrhea and diarrhea index in mice,restore the shape of the ileum of diarrhea model mice,increase the number of goblet cells in the ileum,reduce the levels of IL-6 and TNF-αin the blood,and increase IL-4 and IL-10 and s Ig A content.The experiment indicates that mongolian veterinary drug BSEIC compound can improve the intestinal barrier damage of mice caused by E.coli O8.
