摘 要:试验旨在筛选新疆2个地方绵羊品种在长期自然选择和人工选择过程中受到强烈选择的繁殖性状及脂肪沉积相关候选基因,为绵羊重要经济性状的分子选育奠定基础。对策勒黑羊、吐鲁番黑羊进行全基因组重测序,应用选择信号检测(Fst、Pi),分别在策勒黑羊、吐鲁番黑羊基因组鉴定到受选择的128252个SNPs和128396个SNPs,求交集后进行基因注释,获得63个候选基因,其中与绵羊繁殖相关基因15个(BCL2L11、DDRGK1、AMH、UBE2B、TBX3等);脂肪代谢相关基因8个(IRX3、HOXC10、PNPLA7、PLA2G10等)。这些基因主要富集在视黄醇代谢、TGF-β信号通路、cAMP信号通路、PI3K-Akt信号通路、抗EGFR酪氨酸激酶抑制剂通路等。以上发现为掌握策勒黑羊、吐鲁番黑羊高繁殖力、脂肪沉积等重要经济性状相关功能基因研究及其种质资源保护和利用提供参考依据。In order to screen the candidate genes related to reproductive traits and fat deposits that were strongly selected during long-term natural selection and artificial selection in 2 Xinjiang local sheep breeds,so as to lay a foundation for molecular breeding of important economic traits in sheep.Whole genome sequencing and selection signal detection(Fst,Pi)were performed for Cele black sheep and Turpan black sheep,128252 SNPs and 128396 SNPs which were selected were identified in the genomes respectively.After the intersection,gene annotation was carried out,a total of 63 candidate genes were obtained,including 15 genes related to sheep reproduction(BCL2L11,DDRGK1,AMH,UBE2B,TBX3,etc.),8 genes related to fat metabolism(IRX3 HOXC10 PNPLA7,PLA2G10,etc.).These genes were mainly enriched in retinol metabolism,TGF-beta signaling pathway,cAMP signaling pathway,PI3K-Akt signaling pathway,anti EGFR tyrosine kinase inhibitor resistance,etc.These findings would provide reference for the study of functional genes related to important economic traits,such as high fertility and fat deposition,as well as for the protection and utilization of breed genetic resources of Cele black sheep and Turpan black sheep.
