摘  要:为探讨丁香酚微乳(Eugenol microemulsion,Em)对鸽源金黄色葡萄球菌(S.aureus)临床分离耐药株的抗菌活性及与抗生素联合的作用效果。采用MALDI质谱分析法鉴定从病鸽分离的菌株类别,以ATCC29213为质控菌株,微量肉汤稀释法测定Em和6种抗生素对临床分离株的最低抑菌浓度(MICs),微量棋盘稀释法测定Em与6种抗生素联合作用的分级抑菌浓度指数(FICI),以平板计数法检测Em及其联合多西环素(doxycycline,DOX)对临床分离S.aureus耐药株生长的抑制作用。检测结果表明,分离自病鸽心脏的S.aureus(200113-J3-H1)为环丙沙星(CIP)、黏菌素(COL)、DOX和土霉素(OXY)的多重耐药菌株;Em对该菌的MIC为0.25%,与DOX、OXY、COL和CIP联合作用时均呈现协同作用(FICI=0.125~0.281),其中对DOX的增敏作用最明显,可使其MIC降低16倍;与ENR联合则表现为相加作用(FICI=0.561)。此外,从病鸽肝脏分离的E.coli(200113-J2-L5)为四环素类DOX和OXY耐药菌株;沙门菌(200113-J1-H3)CIP、恩诺沙星(ENR)、卡那霉素(KAN)、COL、DOX和OXY的敏感株,Em对这两种菌的MIC均为0.03125%。结论表明,Em对耐药菌和非耐药菌均具有显著的抑菌活性,有望成为抗生素替代药物,且可显著增强S.aureus多重耐药菌对抗生素的敏感性,这为鸽场细菌性疾病的控制提供了参考。To investigate the antibacterial activity of Eugenol micro-emulsion(Em)and the effect of Em combined with antibiotics against clinically isolated drug-resistant strains of S.aureus from pigeon.The strains isolated from diseased pigeons were identified by MALDI mass spectrometry.The minimal inhibitory concentration(MIC)of Em and six kinds of antibiotics to clinical isolates were determined by microbroth dilution method with ATCC29213 as quality control strain.Micro-chessboard dilution method was used to determine the graded inhibitory concentration index(FICI)of the combined action of Em and six kinds of antibiotics.Plate counting method was used to detect the inhibitory effect of Em and its combination with doxycycline(DOX)on the growth cruve of clinically isolated S.aureus resistant strains.The results showed that S.aureus(200113-J3-H1)isolated from the heart of diseased pigeons was multiple drug resistant strains of ciprofloxacin(CIP),colistin(COL),doxycycline(DOX)and oxytetracycline(OXY).The MIC of Em to this strain was 0.25%,which showed a synergistic effect(FICI=0.125~0.281)when combined with DOX,OXY,COL and CIP,among which the sensitizing effect on DOX was the most obvious and the MIC of DOX could be reduced by 16 times.The combination with ENR showed additive effect(FICI=0.561).In addition,E.coli(200113-J2-L5)isolated from the liver of diseased pigeons was DOX and OXY resistant strains.Salmonella(200113-J1-H3)sensitive strains of CIP,Enrofloxacin(ENR),Kanamycin(KAN),COL,DOX and OXY.The MICs of Em to both bacteria were 0.03125%.Conclusion:Em is expected to become an antibiotic replacement drug,which has significant antibacterial activity against both resistant and non-resistant bacteria.Em can significantly enhance the sensitivity of S.aureus multi-resistant bacteria to antibiotics,simultaneously.This study provides a reference for the control of bacterial diseases in the pigeon house.
