摘  要:目的基于网络药理学的研究方法,探讨黄芪-桂枝药对治疗类风湿关节炎的作用机制。方法通过中药系统药理学数据库与分析平台(TCMSP)及GeneCards、OMIM等数据库搜索药物、疾病的共同作用靶点,制作韦恩图;利用String数据库、Cytoscape 3.6.1软件构建药物疾病共同靶点蛋白互作网络、药物-成分-疾病-靶点网络;最后通过DAVID数据库对共同靶点进行基因本体(gene ontology,GO)、京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)富集分析,并将富集结果可视化。结果黄芪-桂枝药对治疗类风湿关节炎的主要成分是槲皮素、山柰酚、刺芒柄花素、7-O-甲基异丁香酚、β-谷甾醇等;关键作用靶点为TNF、IL6、JUN、IL1B、 CCL2、CXCL8、IL10、ICAM1、MAPK1、PTGS2等;关键作用通路为TNF信号通路、Toll样受体信号通路、HIF-1信号通路、T细胞受体信号通路、细胞凋亡、PI3K-Akt信号通路等。结论黄芪-桂枝药物有效成分通过多靶点、多通路产生抑制炎症反应、免疫功能调节、滑膜血管增殖、细胞凋亡等作用治疗类风湿关节炎。Objective To explore the active mechanism of"Huangqi(Astragali Radix)-Guizhi(Cinnamomi Ramulus)"in the treatment of rheumatoid arthritis based on the research methods of network pharmacology. Methods TCMSP,GeneCards,OMIM databases were used to search for the common targets of drugs and diseases,and the Venn diagram was made. String database and Cytoscape 3.6.1 software were used constructsdrug"disease common target protein interaction"network and"drug-componentdisease-target "network. Finally,GO and KEGG enrichment analysis of common target was carried out through David database,and the enrichment results were visualized. Results The main components of"Huangqi(Astragali Radix)-Guizhi(Cinnamomi Ramulus)"were quercetin,kaempferol,echinonetin,7-o-methylisoeugenol,β-sitosterol,etc;the key targets were TNF,IL6,JUN,IL1 B,CCL2,CXCL8,IL10,ICAM1,MAPK1,PTGS2,etc.;The key pathways were TNF signaling pathway,Toll like receptor signaling pathway,HIF-1 signaling pathway,T cell receptor signaling pathway,apoptosis,PI3 K-Akt signaling pathway. Conclusion The effective components of"Huangqi(Astragali Radix)-Guizhi(Cinnamomi Ramulus)"can inhibit inflammatory reaction,regulate immune function,proliferation of synovial blood vessels and apoptosis through multiple targets and pathways in the treatment of rheumatoid arthritis.
