摘 要:目的通过对脾阳虚大鼠肝组织的代谢组学分析,研究脾阳虚对肝组织代谢的影响。方法 SD大鼠分为正常组和脾阳虚组。其中脾阳虚组采用以小承气汤为基础的多因素法制备模型,并进行脾虚表观指标、体质量曲线、肠吸收功能、脾脏指数和胸腺指数等评估。以非靶向代谢组学分析法定量分析脾阳虚大鼠肝组织的差异表达代谢物,并进行京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路富集分析。结果脾阳虚组大鼠出现明显的脾阳虚证的证候学表现,如体质量降低、毛发稀少、大便湿烂、脾脏指数和胸腺指数降低、肠吸收功能减退等。进一步对脾阳虚大鼠的肝组织进行代谢组学分析,筛选出61个差异表达代谢物(P<0.05),其中26个代谢物降低,35个代谢物升高。KEGG通路富集分析表明,在脾阳虚大鼠肝组织中ABC transporters、Pyrimidine metabolism、Choline metabolism in cancer、Arginine biosynthesis和Central carbon metabolism in cancer等20个重要通路发生显著变化。结论脾阳虚证大鼠肝组织的糖、脂、氨基酸、核酸等代谢均出现显著改变,差异表达的代谢物有助于揭示脾阳虚对肝的影响。Objective To study the effects of spleen Yang deficiency on hepatic metabolism in rats by metabolomics analysis. Methods Male Sprague-Dawley rats were randomly divided into control group and spleen Yang deficiency(SYD)model group. SYD model was made by means of combined factors including starvation,exhaustion and treatment with Xiaochengqi Decoction(小承气汤). And the integrals of SYD,body weight of rats,intestinal absorptive function,spleen index and thymus index were measured to assess spleen Yang deficiency. Furthermore,liver metabolomics was measured to separate liver metabolite profiles of the control group and the SYD model group by ultra-high performance liquid chromatography system coupled with an AB Triple TOF 6600 System. And the Kyoto Encyclopedia of Genes and Genomes(KEGG)was used for metabolic pathways analysis. Results The SYD model group had obvious symptoms of SYD such as losing weight,sparse hairs,mucous stool,decreasing spleen index,decreasing thymus index and decreasing intestinal absorptive function. Liver metabolomics analysis showed that 61 different metabolites were potential biomarker metabolites for SYD. Especially,26 metabolites decreased in the livers of rats with SYD. And 35 metabolites increased in the livers of rats with SYD. Moreover,twenty metabolic pathways,such as ABC transporters,pyrimidine metabolism,choline metabolism in cancer,arginine biosynthesis,and central carbon metabolism in cancer,were significantly influenced by SYD. Conclusion In the liver of rats with SYD there are significant changes in glycometabolism,lipid metabolism,amino acid metabolism and nucleic acid metabolism. The metabolites by metabolomics analysis are helpful to clarify the effects of spleen Yang deficiency on the liver.
