摘  要:目的:分析寻常型银屑病三种不同中医证型患者外周血单核细胞基因和mi RNA的表达,并进行关联分析。方法:收集寻常型银屑病三种中医证型(血瘀证、血燥证、血热证)患者62例和健康对照人群10人的外周血,检测外周血单核细胞的基因表达谱和mi RNA表达谱,利用DIANA在线工具及其数据库对基因芯片和mi RNA芯片结果进行关联分析。结果:与健康对照组比较,三种证型共有的差异mi RNA为hsa-miR-485-3p,三种证型共有的差异基因为RBMS1和FAM98B,血瘀证存在靶标关系的特有差异mi RNA和基因为159对,血燥证存在靶标关系的特有差异mi RNA和基因为15对,血热证不存在证型特有差异mi RNA。结论:差异mi RNA通过调控其靶基因的表达参与银屑病的发生及中医分型。Objective:To analyze the expression of genes and miRNA in peripheral blood mononuclear cells in psoriasis vulgaris patients with three different TCM syndromes,and to conduct correlation analysis.Methods:Peripheral blood was collected from 62 patients with psoriasis vulgaris of the three TCM syndromes(blood stasis syndrome,blood dryness syndrome and blood heat syndrome)and 10 healthy controls,and then the gene expression profile and miRNA expression profile of peripheral blood mononuclear cells were detected.DIANA online tools and databases were applied to correlate the results of gene chip and miRNA chip.Results:Comparing with the healthy control group,the differential miRNA shared by the three syndromes was hsa-miR-485-3 p,and the differential genes shared by the three syndromes were RBMS1 and FAM98 B.There are 159 pairs of miRNAs and genes with a specific target relationship in blood stasis syndrome,and 15 pairs of miRNAs and genes with a target relationship in blood dryness syndrome.There was no miRNAs and genes with a specific target relationship in blood heat syndrome.Conclusion:The differential miRNAs participate in the occurrence of psoriasis and TCM syndromes by regulating the expression of their target genes.
