高效液相色谱法测定温阳活血软胶囊中7种成分的含量

摘　　要：目的建立高效液相色谱法测定温阳活血软胶囊(由红参、赤芍、薤白等组成)中人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1、氧化芍药苷、芍药内酯苷、芍药苷、苯甲酰芍药苷含量的方法。方法采用HPLC-UV-ELSD法测定,色谱柱Agilent TC-C18(4.6 mm×250 mm, 5μm)。人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1的色谱条件为流动相乙腈-水,梯度洗脱,ELSD检测器,漂移管温度60℃,雾化气体为高纯氮气,氮气压力3.0 bar, gain 6;氧化芍药苷、芍药内酯苷、芍药苷、苯甲酰、芍药苷的色谱条件为流动相乙腈-0.1%磷酸水溶液,梯度洗脱,检测波长280 nm,流速1.0 mL/min,柱温30℃。结果人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1、氧化芍药苷、芍药内酯苷、芍药苷、苯甲酰芍药苷分别在0.36~9.00μg(r=0.9996)、0.17~4.41μg(r=0.9997)、0.29~7.25μg(r=0.9997)、0.05~1.25μg(r=0.9997)、0.03~0.72μg(r=0.9999)、0.11~2.72μg(r=0.9997)和0.02~0.48μg(r=0.9999)范围内线性关系良好;人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1、氧化芍药苷、芍药内酯苷、芍药苷、苯甲酰芍药苷平均回收率分别为97.53%、97.45%、98.02%、97.19%、96.98%、97.71%和98.28%,RSD分别为1.26%、1.68%、1.47%、1.67%、1.70%、1.14%和2.72%。结论该方法准确、操作简便、灵敏度高,重复性好,可有效控制温阳活血软胶囊的质量。Objective An HPLC method was established for the determination of ginsenoside Rg1,Re, Rb1 and oxypaeoniflorin, albiflorin, paeoniflorin, benzoylpaeoniflorin in Wenyang Huoxue Soft Capsule(温阳活血软胶囊).Methods An Agilent TC-C18(4.6 mm×250 mm, 5 μm)column was used by HPLC-UV-ELSD.To determine ginsenoside Rg1,Re, Rb1,an ELSD detector was applied.The mobile phase was acrylonitrile–water with gradient elution.The temperature of drift chamber was 60 ℃,and the atomizationgas was nitrogen with 3.0 bar pressure, gain 6.To determine oxypaeoniflorin, albiflorin, paeoniflorin, and benzoylpaeoniflorin, the mobile phase was acetonitrile-0.1%phosphoric acid water with gradient elution.The detection wavelength was 280 nm, the flow rate was 1.0 mL/min, and the column temperature was 30 ℃.Results The calibration curves, linearity and average recovery of the methods were respectively listed as below: 0.36-9.00 μg(r=0.9996),97.53% with RSD of 1.26% in ginsenoside Rg1;0.17-4.41 μg(r=0.9997),97.45% with RSD of 1.68% in ginsenoside Re;0.29-7.25 μg(r=0.9997),98.02% with RSD of 1.47% in ginsenoside Rb1;0.05-1.25 μg(r=0.9997),97.19% with RSD of 1.67% in oxypaeoniflorin;0.03-0.72 μg(r=0.9999),96.98% with RSD of 1.70% in albiflorin;0.11-2.72 μg(r=0.9997),97.71% with RSD of 1.14% in paeoniflorin;0.02-0.48 μg(r=0.9999),98.28% with RSD of 2.72% in benzoylpaeoniflorin.Conclusions The methods are simple and accurate with a high sensitivity and a good repeatability, and so they could be applied to the equality control of Wenyang Huoxue Soft Capsule.
