摘 要:目的观察艾灸关元穴对Wister大鼠衰老进程肝脏组织中SIRT1、FOXO3a、PPARα表达及血清中炎症因子的影响,探讨艾灸关元穴延缓衰老的机制。方法选取SPF级Wistar雌性大鼠18只,其中3月龄6只,为青年组,12月龄大鼠12只,随机分为对照组和艾灸组。艾灸组温和灸关元穴,20 min/d,共40 d。青年组与对照组空白抓取。40 d后,采用酶联合吸附法(ELISA)法检测各组大鼠肝脏组织中的SIRT1、FOXO3a、PPARα的表达水平,血清炎性因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF-α)和白细胞介素-10(IL-10)的表达水平。结果与青年组比较,对照组肝脏组织中SIRT1、FOXO3a、PPARα表达水平明显下降(P<0.01),血清中IL-1β、IL-6、TNF-α、IL-10含量显著上升(P<0.01);与对照组比较,艾灸组肝脏组织中SIRT1、FOXO3a、PPARα表达水平上升(P<0.05),血清中IL-1β、IL-6、TNF-α、IL-10含量显著下降(P<0.01)。结论艾灸关元穴可延缓衰老,其机制可能是激活机体内长寿基因"SIRT1",上调FOXO3a、PPARα的信号通路,引发一系列的抗炎反应进而达到延缓衰老的目的。Objective To observe the effect of moxibustion at Guanyuan(RN4) on the expressions of SIRT1,FOXO3 a and PPARα in liver tissue and the inflammatory factors in serum of aging Wister rats and explore the mechanism of moxibustion at Guanyuan(RN4) to delay aging.Methods A total of 18 SPF-level Wistar female rats were selected, including 6 of them at 3 months old, as the young group, 12 rats at 12 months old, which were randomly divided into the control group and the heath moxibustion group.The moxibustion group was treated with mild moxibustion at Guanyuan(RN4),20 min/day, a total of 40 days.The young group and the control group were randomly crabbed.After 40 days, the expressions of SIRT1,FOXO3 a and PPARα in liver tissue and serum inflammatory factors [interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor(TNF-α) and interleukin-10(IL-10)] of each rats were detected by enzyme-linked immunosorbent assay(ELISA).Results Compared with those of the young group, the expressions of SIRT1,FOXO3 a and PPARα in the liver tissue were significantly decreased(P<0.01),and the levels of IL-1β,IL-6,TNF-α and IL-10 in the serum of the control group were significantly increased(P<0.01).Compared with those of the control group, the expressions of SIRT1,FOXO3 a and PPARα in the liver tissue were increased(P<0.05)and the levels of IL-1β,IL-6,TNF-α and IL-10 in the serum of the health moxibustion group were significantly decreased(P<0.01).Conclusion Moxibustion at Guanyuan(RN4) can delay aging.The mechanism may be activating the longevity gene SIRT1 in the body, up-regulating the signal pathways of FOXO3 a and PPARα and triggering a series of anti-inflammatory reactions, and thereby delaying aging.
