摘  要:目的观察车前子对腹泻大鼠结肠组织AQP3基因蛋白表达的影响,探讨其治疗腹泻的作用机制。方法采用番泻叶灌胃复制腹泻大鼠模型。将60只SD雄性大鼠随机分为正常对照组、模型对照组、阳性药(氢氯噻嗪)对照组和车前子低、中、高剂量组。除正常组外,其余各组均于每日上午灌胃番泻叶药液,正常组灌胃等量蒸馏水;每日下午各治疗组灌服相应药物,正常组和模型组灌服等量的蒸馏水,连续14 d。实验结束后,根据粪便性状和大便次数,统计对比各组大鼠的稀便率、平均稀便级和腹泻指数,采集结肠组织,苏木精-伊红(HE)染色法观察结肠黏膜病理形态学变化,免疫荧光法检测结肠中AQP3蛋白表达情况,qRT-PCR法和Western blot法检测结肠组织AQP3基因和蛋白的表达水平。结果模型组大鼠稀便率、平均稀便级和腹泻指数均明显增加(P<0.01);结肠黏膜表皮细胞出现凋亡坏死,固有层毛细血管扩张充血,少量中性粒细胞浸润;AQP3基因和蛋白表达水平均明显下降(P<0.01)。与模型组比,车前子各剂量组大鼠的稀便率、平均稀便级和腹泻指数均明显降低(P<0.01);车前子各剂量组结肠黏膜表皮细胞凋亡坏死、毛细血管扩张充血和中性粒细胞浸润现象均有改善,以高剂量组改善较为显著;AQP3基因和蛋白表达水平显著升高(P<0.01)。结论车前子具有较好的止泻作用,其作用机制可能与改善结肠黏膜炎性损伤,上调AQP3基因蛋白的表达,提高结肠对水的吸收能力有关。Objective To explore the mechanism of Cheqianzi(Plantaginis Semen)treating diarrhea by observing the effects on expressions of AQP3 gene and protein in colon tissue of diarrhea rats.Methods Fanxieye(Sennae Folium)was used to trigger the diarrhea rats by gavage.Sixty Sprague-Dawley male rats were randomly divided into normal group, model group, positive drug(hydrochlorothiazide) control group, and low, middle and high dose groups of Cheqianzi(Plantaginis Semen).Intragastric administration of Fanxieye(Sennae Folium)was implemented in the morning for 5 groups except the normal group which was implemented by the same dose of distilled water.In the afternoon, each treatment group was given the corresponding drugs, while the normal group and model group were given the same dose of distilled water.The loose stool rate, average ranking of loose stooland diarrhea index were compared according to the fecal traits and stool times after 14 days of treatment.The colon tissues were collected.Hematoxylin-eosin(HE) staining was used to observe the pathological morphological changes of colon tissue.The immunofluorescence method was used to detect of AQP3 protein expression in colon tissue.The qRT-PCR and Western blot methods were used to detect the expressions of AQP3 gene and protein.Results In the model group, the loose stool rate, average ranking of loose stool, and diarrhea index were significantly increased(P<0.01).The epidermal cells of colonic mucosa showed apoptosis and necrosis, telangiectasia and congestion were seen in lamina propriaand a few neutrophils infiltration.The expressions of AQP3 gene and protein significantly decreased(P<0.01).Compared with the model group, the loose stool rate, average ranking of loose stooland diarrhea index were significantly decreased in each dose of Cheqianzi(Plantaginis Semen) groups(P<0.01).The apoptosis and necrosis of colonic mucosal epidermis, telangiectasia and hyperemia and neutrophil infiltration in colonic mucosa were obvious improved in each dose groups, especially in high do
