摘  要:哺乳动物的早期胚胎发育对胚胎着床以及胎儿的长期生长发育都具有决定性作用。酒精对胚胎发育可以产生严重影响,但现有研究主要集中于产前胎儿酒精暴露,对早期胚胎(植入前)酒精暴露关注较少。为进一步阐明酒精导致的胚胎的发育异常机理,本研究利用体外胚胎培养模型,重点研究了酒精暴露对囊胚细胞谱系的分化和胚胎植入的影响。通过对谱系分化标志性分子Cdx2和Oct-4的表达分析及酒精暴露后胚胎着床的分析,发现早期胚胎酒精暴露将严重影响囊胚的谱系分化,并损害胚胎的植入后发育和胎盘的形成状态。结果表明,早期胚胎酒精暴露会打破谱系分化,并严重影响胚胎植入后的宫内发育。The early embryonic development of mammals plays a decisive role in embryo implantation and the long-term growth and development of the fetus.Alcohol can have a serious effect on embryonic development,but existing studies mainly focus on prenatal fetal alcohol exposure and pay less attention to early embryo(preimplantation)alcohol exposure.In order to further clarify the mechanism of abnormal embryo development induced by alcohol,the effects of alcohol exposure on blastocyst cell lineage differentiation and embryo implantation were studied by using the model of embryo culture in vitro.Through the analysis of the expression of Cdx2 and Oct-4,which are the landmarks of pedigree differentiation,and the analysis of embryo implantation after alcohol exposure,it was found that early embryonic alcohol exposure would seriously affect the pedigree differentiation of blastocyst and damage the post-implantation development of embryo and the formation of placenta.The results showed that early embryonic alcohol exposure would break the pedigree differentiation and seriously affect the post-implantation development in uterus of the embryo.
