摘 要:目的基于16SrRNA测序分析肾结石患者结石及尿液微生态结构。方法选取2016年3月至2020年3月本院收治的肾结石患者102例为研究对象,均行经皮肾镜取石术。采用16SrRNA测序技术检测结石标本和尿液标本中的微生物信息,根据结石成分将标本分为碳酸磷灰石组、草酸钙组、磷酸铵镁组、尿酸组,分析各组样本菌群的多样性和丰富度。结果结石标本中菌群门水平以变形菌门、厚壁菌门、放线菌门为主,科水平以肠杆菌科、黄单胞菌科、假单胞菌科为主,属水平以寡养单胞菌属、假单胞菌属、短波单胞菌属为主;变形菌门是四组结石标本中的优势菌门(>55%),磷酸铵镁组中放线菌门和无壁菌门是仅次于变形菌门的优势菌(>8%),碳酸磷灰石组、草酸钙组中厚壁菌门和放线菌门是仅次于变形菌门的优势菌(>8%),尿酸组中拟杆菌门和放线菌门是仅次于变形菌门的优势菌(>5%);在属水平中,磷酸铵镁组中的优势菌是奇异变形杆菌、寡养单胞菌属、棒状杆菌属,碳酸磷灰石组是假单胞菌属、寡养单胞菌属、金黄杆菌属,草酸钙组是假单胞菌属、寡养单胞菌属、短波单胞菌属,尿酸组是寡养单胞菌属、假单胞菌属、金黄杆菌属。尿液标本中菌群门水平以变形菌门、厚壁菌门、无壁菌门为优势菌(>13%),属水平以假单胞菌属、支原体属、克雷伯氏菌属为优势菌(>8%);结石标本和尿液标本α多样性指标比较,差异无统计学意义(P>0.05):结论肾结石患者结石标本中寡养单胞菌、金黄杆菌和短波单胞菌含量较高,而尿液中则以假单胞菌属、支原体菌属、乳酸杆菌属为优势菌。Objective To investigate the urinarv microbiome and stone microbiome of kidney stone patients based on 16SrRNA sequencing analysis.Methods From March 2016 to Marc h 2020,102 patients with kidney stone underwent percutaneous nephrolithotomy in our hospital were selected.The urinary microbiome and stone microbiome were analyzed through 16SrRNA sequencing analysis.Then the diversity and abundance of bacteria were also analyzed.Results In stone samples,pro-teohacteria,phylumfirmicutes and actinobacteria were dominant at the phylum level.Enterobacteriace-ae,xanthomonadaceae and pseudomonadaceae were dominant at the family level,and stenotroph-omonas,pseudomonas and brevundimonas were dominant at the genus level.Proteobacteria was the dominant phylum(>55%)among four groups of stone specimens.Except proteobacteria,actinomy-cetes and tenericute were the dominant bacteria in magnesium ammonium phosphate group(>8%),phylum fimiicutes and actinobacteria were the dominant bacteria in carbonate apatite group and calcium oxalate group(>8%),bacteroidetes and actinobacteria were the dominant bacteria in uric acid group(>5%).The dominant bacteria in magnesium ammonium phosphate group were proteusmirabilis,stenotrophomonas and corynebacterium.The dominant liacteria in c arbonate apatite group were pseudomonas,stenotrophomonas and chryseobacterium.The dominant bacteria in calcium oxalate group were pseudomonas,stenotrophomonas and brevundimonas.The dominant bacteria in uric acitl group were stenotrophomonas,pseudomonas and chryseobacterium.In urine samples,proteobacteria,phylum firm-icutes and tenericute were the dominant bacteria(>13%),and pseudomonas,mycoplasma and kleb-siella were dominant bacteria(>8%)at genus level.There was no significant difference in alpha diversity between stone samples and urine samples(P>0.05).Conclusions The diversity between stone and urine microbiome of patients with kidney stone has no significant difference,but bacterial species have obvious difference between two samples.Stenotrophomonas,chryseobact
