摘  要:为探究高压均质(HPH)处理鸡蛋卵黄对猪精液冷冻效果的影响。选取Tris—柠檬酸—葡萄糖基础稀释液,以未处理的鸡蛋卵黄(20%EY)为对照组,添加经HPH处理后的鸡蛋卵黄(20%EY)为试验组,制成Ⅰ液和Ⅱ液,冷冻-解冻后分别对猪精子活力、活率、顶体完整率、质膜完整率、线粒体活性、抗氧化指标及DNA完整率和凋亡指标进行检测。结果显示,经HPH处理能有效提高猪精子冻后质量。精液解冻后精子活力、活率、顶体完整率、质膜完整率分别为84.32%、86.71%、68.92%、59.09%,较对照组分别提高了13.7%、16.58%、6.5%、4.88%(P<0.05)。与对照组相比,试验组的MDA和ROS较对照组均有所上升(P>0.05),SOD较对照组有所下降(P>0.05)。但冻后精子NO和NOS含量显著降低(P<0.05)。试验组的精子解冻后DNA片段完整性较对照组明显提高(P<0.05),凋亡率下降(P<0.05)。结果表明,HPH处理鸡蛋卵黄能有效提高猪精液冷冻后质量。The aim of this study was to explore the effect of egg yolk(EY)treated by high pressure homogenization(HPH)on the boar semen freezing.Ejaculates were collected from five healthy,mature Duroc boars.Based on the Tris-citric acid-glucose(TCG)diluent,untreated 20%EY was used as the control group,and the HPH-EY was used as the treated group.Frozen semen samples were thawed and assessed for sperm motility,viability,plasma acrosome integrity,membrane integrity,mitochondrial activity,and DNA integrity,reactive oxygen species(ROS),superoxide dismutase(SOD),malondialdehyde(MDA).Our results revealed the HPH-EY improved the quality of post-thawed sperm after cryopreservation.The semen extended in the HPH-EY extract tended to have a higher sperm viability(84.32%),motility(86.71%),acrosome integrity(68.92%),and membrane integrity(59.09%)than the control group(P<0.05).The extenders supplemented HPH-EY significantly increased the MDA and ROS activities(P<0.05),and decreased the SOD activity and the concentrations of NO and NOS compared to the control group(P<0.05).The extenders supplemented HPH-EY significantly increased DNA integrity(P<0.05),and decreased the apoptotic rate compared with controls(P<0.05).The present study showed that adding HPH-EY to the semen extender enhanced the post-thaw qualities of boar semen.
