摘  要:长链酰基辅酶A合成酶(LACS)是脂肪代谢网络中的关键酶,催化长链酰基辅酶A的合成并在多种生物过程中发挥作用。本研究以拟南芥中长链酰基辅酶A合成酶家族的基因序列为参考来探究LACS在陆地棉基因组中的数量和分布情况。通过生物信息学方法综合分析陆地棉GhLACS基因家族,发现该家族含有21个成员,CDS序列长度为1938-2274 bp,编码氨基酸数量为645-757个,定位在15条染色体上。SOPMA预测蛋白二级结构,发现该家族成员的蛋白二级结构均以α-螺旋、β-转角、延伸链、无规则卷曲组成。亚细胞定位分析发现7个基因定位在过氧化物酶体,6个基因定位在叶绿体,8个基因定位在内质网。基因结构分析表明同一亚组内含子和外显子的排列分布较为相似,而不同亚组间存在一定的差异性。本研究有助于了解陆地棉Gh LACS家族的功能,并且为棉籽油脂质代谢提供理论依据。Long-chain acyl-CoA synthetase(LACS)is a key enzyme in the fat metabolism network,catalyzing the synthesis of long-chain acyl-CoA and playing an important role in various biological processes.This study was based on the gene sequence of the long-chain acyl-CoA synthetase family in Arabidopsis to explore the quantity and distribution of LACS in the genome of Gossypium hirsutum L..The GhLACS gene family of Gossypium hirsutum L.was comprehensively analyzed by bioinformatics.It was found that the family contained 21 members,the CDS sequence length is 1938~2274 bp,the number of amino acids encoded was 645~757,and which was located on 15 chromosomes.By SOPMA predicted protein secondary structure,it was showed that the protein secondary structure of the family members were composed ofα-helix,β-turn,extended chain,and random coil.Subcellular localization analysis revealed that 7 genes were located in peroxisomes,6 genes in chloroplasts,and 8 genes in the endoplasmic reticulum.Gene structure analysis confirmed that the distribution of introns and exons in the same subgroup was similar,while there were some differences among different subgroups.This study is helpful to understand the function of the GhLACS family of Gossypium hirsutum L.and provides a theoretical basis for the lipid metabolism of cottonseed oil.
