摘  要:蔗糖磷酸合成酶(SPS)和磷酸蔗糖磷酸酶(SPP)是植物蔗糖合成的重要酶,在调控蔗糖合成及其积累等方面有重要作用。辣椒基因组测序的完成及转录组数据的公布为鉴定和探究辣椒SPS和SPP基因的多样性提供了机会。本研究利用生物信息学方法对辣椒SPS和SPP基因进行鉴定,并从染色体定位、系统发育关系和表达模式分析其特征。结果发现,两个辣椒共含有4个SPS和SPP基因,每个辣椒种均编码2个成员,SPS编码的氨基酸长度和分子量约是SPP的2倍,等电点范围6.12-8.16和5.96-6.26之间。染色体定位发现辣椒SPS和SPP基因均定位在不同的染色体上。系统发育树的构建来自7个物种,结果表明SPS和SPP都可以分为4个亚族。表达谱分析显示,基因呈现差异表达,其中Ca-LZSPS1和Ca-LZSPP2呈现广谱性表达;进一步研究发现,SPS和SPP基因都受到3种激素(ABA,IAA,SA)不同程度的诱导;此外,在逆境胁迫下,辣椒SPS相比SPP更容易受到环境诱导,而SPP基因则在叶中主要参与热调控机制,在根中更多的参与冷调控机制。这些结果阐明了辣椒SPS和SPP基因的表达特性,为进一步研究辣椒中蔗糖的合成与积累提供理论依据。Sucrose phosphate synthase(SPS)and sucrose-6-phosphate phosphatase(SPP)are important enzymes in plant sucrose synthesis,such as regulating sucrose synthesis and accumulation.The publication of pepper genome and transcriptome data provides an opportunity to explore and identify the diversity of pepper SPS and SPP gene.This study identified SPS and SPP genes and the characteristics of their chromosome location,phylogenetic tree relationship and expression patterns were analyzed by bioinformatics methods.The results showed that two pepper contains four SPS and SPP genes respectively,each pepper species was encoding two members.SPS encoding amino acids of the length and the molecular weight is about double the SPP,and the isoelectric point range between 6.12~8.16 and 5.96~6.26 respectively.Chromosomal localization found that these genes were distributed on different chromosomes,and Ca-LZSPP2’s chromosome location was uncertain.The phylogenetic tree of SPS and SPP genes were built from seven species and divided into four subgroups respectively.Expression profile analysis showed that genes were differentially expressed,in which CA-LZSPS1 and CA-LZSPP2 were widely expressed.Further studies revealed that SPS and SPP genes were induced to different degrees by three hormones(ABA,IAA,SA).In addition,SPS is more susceptible to environmental induction than SPP under adversity stress,and SPP gene is mainly involved in the heat regulation mechanism in leaves and the cold regulation mechanism in roots.These results provide data basis for further study of SPS and SPP genes in pepper.
