摘 要:以二年生膜荚黄芪为试验材料,探究生态因子对膜荚黄芪根中黄芪甲苷含量及其关键酶基因表达量的影响。测定二年生黄芪的黄芪甲苷含量,并采用实时荧光定量PCR分析黄芪甲苷合成途径中7个关键酶:乙酰辅酶A乙酰基转移酶(AACT)、甲羟戊酸激酶(PMK)、甲羟戊酸焦磷酸脱羧酶(MVD)、异戊二烯焦磷酸异构酶(IDI)、法尼基焦磷酸合酶(FPS)、角鲨烯合酶(SS)、角鲨烯环氧酶(SE)基因表达量。结果表明:光合有效辐射、土壤湿度、温度、降雨量、最大降雨强度均会影响根部黄芪甲苷合成,其中温度和光合有效辐射与黄芪甲苷含量呈显著正相关(P<0.05),土壤湿度、降雨量、最大降雨强度与黄芪甲苷含量呈负相关。黄芪甲苷合成关键酶基因在黄芪甲苷积累的重要时期表达较高,黄芪甲苷合成关键酶基因的表达相互影响,膜荚黄芪根中关键酶基因的表达对黄芪甲苷积累大多为正相关关系;其中PMK基因与黄芪甲苷含量呈显著正相关(P<0.05),为主要调控基因。Biennial Astragahss membranaceus was used as the material to explore the effects of ecological factors on astragaloside content of Astragahss membranaceus and expression quantity of key enzyme genes. Astragaloside content of biennial Astragahss membranaceus was measured, and real-time fluorescent quantitative PCR was applied to analyze gene expression quantity of 7 key enzymes, AACT, PMK, MVD, IDI, FPS, SS and SE, in the synthetic route of astragaloside. The results showed that photosynthetically active radiation, soil humidity, temperature, rainfall and the maximum rainfall intensity could affect astragaloside synthesis in root, of which temperature had a significant negative correlation with the content of astragaloside(P<0.05), photosynthetically active radiation had a significant positive correlation with the content of astragaloside(P<0.05), and soil humidity, rainfall and the maximum rainfall intensity had a negative correlation with the content of astragaloside synthesis. The key enzyme genes of astragaloside had high expression in the important period of astragaloside accumulation. The expression of key enzyme genes of astragaloside synthesis was correlated. Generally, the expression of key enzyme genes in root of Astragahss membranaceus had a positive correlation with the accumulation of astragaloside, where PMK gene had a significant positive correlation with the content of astragaloside(P<0.05), which was the main regulatory gene.
