运用协同戊巴比妥钠模型探讨鲜人参膏对小鼠的促睡眠作用

摘　　要：运用协同戊巴比妥钠模型研究人参加工品"鲜人参膏"(Fresh ginseng paste, FGP)对小鼠睡眠的促进作用。观察小鼠睡眠潜伏期及睡眠时长,并检测小鼠血清中五羟色胺(5-HT)和一氧化氮合酶(NOS)水平,评价小鼠睡眠状况;通过H&E染色法观察小鼠海马体组织病理学变化。结果表明:与戊巴比妥钠组比,FGP协同戊巴比妥钠组小鼠的睡眠潜伏期显著缩短(P<0.05),而睡眠时长显著延长,FGP(400 mg/kg)效果最佳。与正常对照组比,戊巴比妥钠组与给药组小鼠的5-HT与NOS含量显著提升(P<0.05);与戊巴比妥钠组相比,FGP给药组小鼠的5-HT与NOS含量显著提升(P<0.05)。此外,FGP在协同戊巴比妥钠增强镇静安神作用的基础上,降低了戊巴比妥钠对小鼠中枢神经的副作用。因此,FGP具有显著的镇静安神作用,可显著延长小鼠的睡眠时长。In this paper, the effects of fresh ginseng paste(FGP) and pelltobarbitalum natricum(PN) model on sleep in mice were investigated. Sleep latency and sleep duration of mice were observed, and the levels of serotonin(5-HT) and nitric oxide synthase(NOS) in serum of mice were detected to evaluate sleep status of mice. The pathological changes of renal tissues were observed by using H&E staining. The results significantly showed that compared with the PN group, sleep latency of the FGP group was notably shortened(P<0.05), and sleep duration was significantly prolonged, and FGP(400 mg/kg) was the best.Compared with the normal control group, 5-HT and NOS levels of mice in the PN group and the FGP group were significantly increased(P<0.05);compared with the PN group, the levels of 5-HT and NOS in mice receiving FGP were significantly increased(P<0.05). In addition, FGP reduced the side effects of PN on the central nervous system in mice on the basis of enhancing the sedative and soothing effect of PN. Therefore, FGP has a significant calming and soothing effect.
