摘  要:目的:探讨糖肾方对UUO模型小鼠肾脏氧化损伤的影响。方法:选用7周龄的雄性C57BL/6N小鼠,随机分为假手术组、UUO模型组(UUO模型7 d组、UUO模型14 d组)、糖肾方组(UUO+糖肾方7 d组、UUO+糖肾方14 d组)、福辛普利组(UUO+福辛普利7 d组、UUO+福辛普利14 d组)。预给药5 d后造模,术后继续给药至7 d或14 d。观察肾脏病理,检测肾组织和血清MDA含量及SOD活性,并检测肾组织NADPH氧化酶的表达。结果:与假手术组比较,UUO模型小鼠肾间质炎性细胞呈弥漫性浸润,胶原纤维大量增生,糖肾方干预可以减轻UUO小鼠肾间质炎性细胞浸润,减少肾间质胶原纤维增生,降低肾组织和血清MDA含量、升高SOD活性,下调肾组织中Nox4和磷酸化p47phox的表达。结论:糖肾方可以改善UUO小鼠的肾组织病理损伤,减轻肾间质纤维化,机制可能与下调NADPH氧化酶的表达,从而降低UUO小鼠肾脏和血清的氧化应激水平有关。Objective:This study aims to investigate the effect and mechanism of Tangshen Formula(TSF)on improving the renal injury in Unilateral Ureteral Obstruction(UUO)mice by reducing the expression of NADPH oxidase and the oxidative stress in kidney.Methods:Seven-week-old male C57BL/6N mice were randomized into 4 groups:sham surgery group,model group(7-day UUO model group,14-day UUO model group),TSF group(UUO and 7-day TSF group,UUO and 14-day TSF group),fosinopril group(UUO and 7-day fosinopril group,UUO and 14-day fosinopril group).The drug was given for 5 days,then the model was established,and the drug was given for another 7 or 14 days after surgery.The contents of MDA in renal tissue and serum and the activity of SOD were measured,and renal pathological changes were observed.In addition,the expression of NADPH oxidase in renal tissue were detected.Results:Compared with sham surgery group,diffuse infiltration of renal interstitial inflammatory cells and sharp increase in proliferation of collagen fibers were found in mice of UUO model.TSF could reduce such infiltration and proliferation,and decrease the expressions of Nox4 and phosphorylated P47phox,contents of MDA in renal tissue and serum,but increase the activity of SOD.Conclusion:TSF can improve the pathological injury of renal tissue in UUO mice and reduce the renal interstitial fibrosis.The mechanism may be related to the down-regulation of NADPH oxidase expression,thus reducing the oxidative stress in renal tissue and serum of UUO mice.
