摘 要:目的:探讨补阳还五汤对内皮祖细胞功能(EPCs)的干预作用,探索该方的治疗作用机制。方法:本研究取健康大鼠54只,共分为空白组、假手术组、急性心肌梗死大鼠模型组、中药干预组、西药对照组及中西医结合组。对大鼠进行分组干预,空白组给予等量生理盐水灌胃;假手术组假手术后给予等量生理盐水灌胃;模型组造模后给予等量生理盐水灌胃;中药干预组造模后给予补阳还五汤灌胃;西药对照组造模后给予阿托伐他汀灌胃;中西医结合组造模术后给予补阳还五汤和阿托伐他汀钙灌胃。运用密度梯度离心的方法,分离培养各组大鼠EPC,采用四甲基偶氮唑盐微量酶反应比色法、黏附能力测定实验以及改良的Boyden小室的方法,分析对比各组EPC的增殖、黏附以及迁移能力。结果:第7天、第14天及第4周,模型组大鼠外周血EPCs计数、增殖、迁移及黏附能力均较假手术组显著下降;中药干预组、西药对照组及中西医结合组外周血EPCs计数、增殖、迁移及黏附能力较模型组均显著增加,差异有统计学意义(P<0.05);中西医结合组较中药干预组及西药对照组对EPCs功能的改善作用更为显著,差异有统计学意义(P<0.05)。结论:他汀类药物联合补阳还五汤较单纯的中药或西药治疗可更好地改善急性心肌梗死大鼠的内皮功能,为改善急性心肌梗死患者远期预后提供新的研究切入点。Objective:To determine the effect of Buyang Huanwu Decoction on endothelial progenitor cells(EPCs)function,and to explore the therapeutic mechanism of Buyang Huanwu Decoction.Methods:A total of 54 healthy rats were divided in⁃to the blank group,the control group,the model group,the Chinese medicine group,the western medicine group,and the com⁃bined traditional Chinese and western medicine group.Density gradient centrifugation separation was applied to isolate and culture EPCs.Tetramethyl azo salt trace enzyme reaction colorimetry,adhesion ability test and improved Boyden chamber were used to compare the EPCs proliferation,adhesion,and migration among the 6 groups.Results:On day 7,14 and week 4,the model group showed significantly lower peripheral blood EPCs count,and stronger proliferation,migration and adhesion ability com⁃pared with that of the control group;The Chinese medicine group,western medicine control group and the combined traditional Chinese and western medicine group showed significantly higher peripheral blood EPCs count,and stronger ability of prolifera⁃tion,migration and adhesion compared with the model group(P<0.05);The combined traditional Chinese and western medicine group showed more significant improvement in the function of EPCs compared to the Chinese medicine group and the western med⁃icine group(P<0.05).Conclusion:Statins combined with Buyan Huanwu Decoction can better improve the endothelial function of acute myocardial infarction rats and provide a new perspective on improving the long-term prognosis of acute myocardial infarc⁃tion patients.
