摘 要:为了解猪圆环病毒3型(porcine circovirus type 3,PCV3)在云南省的流行及遗传进化情况,2020年10—12月收集云南省部分猪场疑似病料共24份,采用PCR方法进行PCV3病原检测;选取PCV3阳性扩增产物进行克隆测序,并将测序结果与16株国内外参考毒株进行ORF2核苷酸序列同源性分析。结果显示:24份病料样品中检出阳性5份,阳性检出率为20.8%;克隆出的阳性扩增产物(YNJS-2020)ORF2核苷酸序列和国内外参考毒株的同源性较高,为97.4%~98.9%,其中与西班牙毒株MF80572000同源性最高,与安徽14-201611毒株同源性最低。结果表明,云南省存在PCV3流行,且流行毒株与国内外其他毒株遗传关系较近,但也有一定差异。结果提示,云南省应加强对PCV3的监测及其流行的控制,避免PCV3大面积流行。本研究为云南省PCV3基因型分析和疫苗研究提供了一定的数据基础。In order to identify the status of prevalence and genetic evolution of porcine circovirus type 3 PCV3 in Yunnan Province 24 suspected samples were collected from some swine farms during October to December 2020 to detect PCV3 by PCR.PCV3-positive amplified products were cloned and sequenced and the results were compared with 16 domestic and foreign reference strains for ORF2 nucleotide sequence homology analysis.The results showed that 5 out of 24 samples were positive,with a positive detection rate of 20.8%.The ORF2 nucleotide sequence of cloned positive amplified product(YNJS-2020)had high homology with the reference strains(97.4%to 98.9%),and the homology was highest with Spanish PCV3 strain MF80572000 and lowest with Anhui strain 14-201611.It was concluded that PCV3 was prevalent in the province which had a close genetic relationship with other viral strains although there was a certain gap.It was suggested that PCV3 should be intensively monitored and controlled to avoid any spreading on a large scale.A certain data were hereby provided for PCV3 genotype analysis and development of corresponding vaccines in Yunnan Province.
