摘  要:目的:优选归芪通脉合剂的水提工艺,为其后续研究提供参考。方法:以挥发油的提取率为指标,采用单因素试验优选挥发油的提取工艺。以黄芪甲苷、羟基红花黄色素A(HSYA)的含量和干膏得率的综合评分为指标,采用层次分析法(AHP)确定各指标权重系数,通过L 9(34)正交试验考察提取次数、提取时间和料液比,优选水提工艺,并进行验证实验。结果:以AHP确定黄芪甲苷、HSYA、干膏得率的权重系数分别为0.6483、0.2297、0.1220,筛选的提取工艺为当归等含油药材加入10倍量的水,提取5 h,收集蒸馏液,水提液另器保存,滤渣与剩余10味药共水提取2次,首次加入12倍水,第二次提取加入10倍水,每次提取1 h。3次验证实验结果显示,黄芪甲苷、HSYA的平均质量分数分别为0.18%、1.47%,平均出膏率为35.29%,平均综合评分为98.73,RSD为1.20%。结论:AHP确定的权重系数科学、有效,优选的工艺条件稳定可行,适用于规范化生产。Objective:To optimize the water extraction process of Guiqi Tongmai Mixture and thereby lay a basis for the follow-up research.Methods:The extraction process of volatile oil was optimized by single-factor test with the extraction rate as evaluation index.Then,the weight coefficients of indexes astragalosideⅣ,hydroxysafflor yellow A(HSYA),and extract yield were determined by analytic hierarchy process(AHP)and the extraction times,extraction duration,and liquid-to-material ratio were optimized by L 9(34)orthogonal test.In addition,validation test was performed.Results:The weight coefficients of astragalosideⅣ,HSYA,and dry extract yield were 0.6483,0.2297,and 0.1220,respectively.The optimized extraction process is as follows.Angelicae Sinensis Radix and other medicinal materials containing volatile oil are extracted with water(liquid-to-material ratio=10∶1)for 5 h and the distillate is collected.The water extract is stored separately and the filter residue is put together with the remaining 10 medicinal materials for twice extraction(liquid-to-material ratio is 12∶1 for the first time and 10∶1 for the second time),1 h each time.According to the results of three validation tests,the average content of astragalosideⅣand HSYA was 0.18%and 1.47%,respectively,and the average extract yield was 35.29%.The average comprehensive score was 98.73(RSD=1.20%,n=3).Conclusion:The weight coefficients determined by AHP are scientific and effective and the optimized extraction process is stable and feasible,which can be applied to standardized production.
