摘 要:目的对系统性红斑狼疮(SLE)血中的微小核糖核酸(miRNA)和信使RNA(mRNA)相关的基因表达谱进行综合的生物信息学分析。方法拟定检索策略,从数据库中下载与SLE相关的基因表达谱,通过软件/数据库进行差异基因筛选、miRNA靶基因预测、miRNA靶基因与mRNA取交集基因进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)功能富集、蛋白-蛋白互作网络和miRNA-mRNA互作网络。结果共纳入3个数据集,miRNA靶基因与mRNA有720个交集基因,且这些基因富集到CXC趋化因子活性、B细胞分化等重要的功能,以及经典的Wnt、Toll样受体等通路;分析得出蛋白-蛋白和miRNA-mRNA相互作用网络。结论miRNA可能通过直接靶向作用调控靶基因,进而影响SLE患者体内多种基因及信号通路的网络调控,从而参与SLE疾病的发生和发展。Objective To comprehensive bioinformatics analysis of microribonucleic acid(miRNA)and messenger RNA(mRNA)related gene expression profiles in the blood of systemic lupus erythematosus(SLE).Methods Develop a search strategy,download gene expression profiles related to SLE from GEO(Gene Expression Omnibus),perform differential gene screening,miRNA target gene prediction,miRNA target gene and mRNA extraction through software/database for gene ontology(GO)and Kyoto Encyclopedia of Gene and Genome(KEGG)function enrichment,protein-protein interaction network and miRNA-mRNA interaction network.Results A total of 3 data sets are included.There are 720 overlapping genes between miRNA target genes and mRNA,and these genes are enriched for important functions such as CXC chemokine activity,B cell differentiation,and classic Wnt,Toll-like receptor pathways.The analysis results in the protein-protein and miRNA-mRNA interaction network.Conclusion MiRNA may directly regulate target genes through direct targeting,thereby affecting the network regulation of multiple genes and signal pathways in SLE patients,thereby participating in the occurrence and development of SLE diseases.
