摘 要:目的探讨藏红花酸预处理能否减轻心肌梗死大鼠心肌细胞的炎症反应、减轻心肌形态学损伤、保护大鼠心脏功能和维护血流动力学稳定。方法连续2 d静脉注射异丙肾上腺素[90 mg/(kg·d)]建立大鼠心肌梗死模型。将50只大鼠随机分为5组:对照组、心肌梗死组、藏红花酸50 mg/(kg·d)组、藏红花酸100 mg/(kg·d)组、藏红花酸200 mg/(kg·d)组。藏红花酸50 mg/(kg·d)组、藏红花酸100 mg/(kg·d)组、藏红花酸200 mg/(kg·d)组大鼠予以相应剂量藏红花酸灌胃,对照组与心肌梗死组予以等量的生理盐水灌胃,每日1次,连续15 d。在按剂量操作后通过评估左心室功能评价其对心肌的作用。检测心肌组织匀浆中的肌酸激酶(CK-MB)、乳酸脱氢酶(LDH)和炎性细胞因子。通过苏木素-伊红染色进行组织病理学研究。结果与心肌梗死组比较,藏红花酸各剂量组平均动脉压、舒张压、收缩压和心率均提高,并且呈剂量依赖性(P<0.05或P<0.01)。与心肌梗死组相比,藏红花酸各剂量组的组织匀浆中CK-MB、LDH水平增高(P<0.05或P<0.01)。藏红花酸可以使心肌梗死大鼠心肌组织中的炎性细胞因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平明显降低(P<0.05或P<0.01)。藏红花酸可以逆转心肌梗死大鼠心脏组织切片的细胞结构变化。结论藏红花酸可以通过减轻炎症反应,维护心肌细胞膜的稳定性,保护心肌梗死大鼠的心脏功能,维持血流动力学稳定,使其成为心肌梗死的一种可能的保护剂。Objective To investigate whether crocetin pretreatment can reduce the inflammatory response of myocardial cells in rats with myocardial infarction,reduce the morphological damage of myocardium,protect the cardiac function of rats,and maintain the dynamic stability of blood flow.Methods Isoproterenol[90 mg/(kg·d)]was injected intravenously for two consecutive days to establish rat myocardial infarction model.The rats were divided into 5 groups:control group,myocardial infarction group,crocetin 50 mg/(kg·d)group,crocetin 100 mg/(kg·d)group,and crocetin 200 mg/(kg·d)group.The rats in control group and myocardial infarction group were given normal saline,and the rats in crocetin dose groups were given oral doses of 50,100,and 200 mg/(kg·d)of crocetin for 15 days.Creatine kinase(CK-MB),lactate dehydrogenase(LDH),and inflammatory cytokines were detected in myocardial tissue homogenate.Histopathological studies were performed with hematoxylin and eosin staining.Results Compared with myocardial infarction group,mean arterial pressure(MAP),diastolic pressure(DAP),systolic arterial pressure(SAP),and heart rate increased in crocetin treatment group in a dose-dependent manner(P<0.05 or P<0.01),the levels of CK-MB and LDH in the tissue homogenate increased in crocetin treatment group(P<0.05or P<0.01),the levels of inflammatory cytokines interleukin-1β(IL-1β),interleukin-6(IL-6),and TNF-αin myocardial tissue of myocardial infarction rats significantly reduced in the crocetin treatment group(P<0.05 or P<0.01).The changes of cell structure in heart tissue sections of rats with myocardial infarction were reversed in the crocetin treatment group.Conclusion Crocetin can reduce inflammation,maintain the stability of myocardial cell membrane,protect the heart function,and maintain hemodynamic stability of myocardial infarction rats.
