摘  要:【目的】本文利用GFP标记系统研究秸秆还田对假单胞菌PW9土壤定殖能力的影响。【方法】将可产生绿色荧光蛋白(GFP)的质粒pBBR1MCS2-Tac-EGFP电转化至假单胞菌PW9中,通过抗生素鉴定及质粒检测,获得了能够产生稳定的强烈绿色荧光且具有较高解磷能力的标记菌株PW9-gfp,并以此为试验菌株进行土壤定殖能力研究。【结果】标记菌株PW9-GFP施入土壤0~60 d时间内,两种处理方式的土壤样品中有效菌落数量均由109 CFU/g降至10 CFU/g;在植株同一生长时间时,标记菌株PW9-gfp单独施入的土壤样品中有效菌落数量均略低于与秸秆同时施入的土壤样品。【结论】秸秆还田对菌株PW9的土壤定殖能力具有一定的促进作用,其作用机理有待进一步研究,这一研究结果为解磷菌合理施用及微生物菌肥与秸秆还田协同作用深入研究奠定了一定的理论基础。【Objective】The effect of straw mulching on pseudomonas PW9 soil colonization ability was studied by GFP labeling system.【Method】The plasmid PBBR1MCS2-TAC-EGFP,which could produce green fluorescent protein(GFP),was electrotransformed into pseud-omonas PW9,by antibiotic identification and plasmid detection,the labeled strain PW9-GFP was obtained,which could produce stable and strong green fluorescence and had high phosphorus solubility,and the colonization ability of soil was studied with the strain.【Result】The mark strain PW9-GFP was applied to the soil within 0-60 days,the number of effective colonies in the soil samples under both treatments decreased from 109 CFU/g to 10 CFU/g;When the plants were growing at the same time,the number of effective colonies in soil samples treated with mark strain PW9-GFP was slightly lower than that of soil samples treated with straw.【Conclusion】Straw mulching had a certain effect on soil colonization of strain PW9,its mechanism of action needed further study,this results lay a theoretical foundation for the rational application of phosphorus-releasing bacteria and the further study of the synergistic effect of microbial fertilizer and straw mulching.
