苦参碱对TGF-β1诱导的人腹膜间皮细胞成纤维细胞生长因子2的调控作用

摘　　要：目的研究苦参碱对成纤维细胞生长因子2(FGF-2)在人腹膜间皮细胞(HPMCs)上皮间充质转分化(EMT)中的调控作用。方法将HPMCs分为三组(对照组、TGF-β1刺激组及0.8 mg/mL苦参碱+TGF-β1处理组);TGF-β1刺激组用含5 ng/mL TGF-β1的完全培养基,处理组用含0.8 mg/mL苦参碱和5 ng/mL TGF-β1的完全培养基,对照组用等量完全培养基。干预处理48 h后,采用转录组测序分析各组HPMCs细胞FGF-2的mRNA表达情况,最后采用蛋白免疫印迹和实时荧光定量PCR分别检测各组HPMCs细胞FGF-2蛋白和mRNA的表达情况。结果 5 ng/mL TGF-β1刺激HPMCs细胞,发现其能上调FGF-2的蛋白和mRNA的表达水平(其中蛋白上调2.00倍,mRNA上调6.18倍),而苦参碱干预后能显著下调TGF-β1刺激后FGF-2的蛋白和mRNA的表达水平(其中蛋白下调1.77倍,mRNA下调4.30倍)。结论苦参碱通过对成纤维细胞生长因子2发挥抑制作用,进而阻止成纤维细胞在腹膜聚集,最终防止腹膜纤维化的发生。Objective To investigate the regulatory effect of matrine on fibroblast growth factor 2(FGF-2)during epithe-lial mesenchymal transition(EMT)of peritoneal mesenchymal cells.Methods Human peritoneal mesothelial cells were divided into three groups according to different treatment methods(the control group,the TGF-β1 stimulation group and the TGF-β1+0.8 mg/mL matrine treatment group).The complete medium containing 5 ng/mL TGF-β1 was used in the TGF-β1 stimulation group,and the complete medium containing 5 ng/mL TGF-β1 and 0.8 mg/mL matrine was used in the treatment group.The control group was completely cultured with the same amount,and after 48 h of inter-vention treatment,the mRNA expression level of FGF-2 was analyzed by mRNA-seq.Finally,the mRNA and protein expression levels of FGF-2 in human peritoneal mesothelial cells were detected by relative real-time fluorescence quantitative PCR and western blot.Results It was found that 5 ng/mL of TGF-β1 could up-regulate the expression levels of FGF-2 mRNA and protein when stimulating human peritoneal mesothelial cells(6.18 times mRNA and 2.00 times protein).After matrine intervention,the mRNA and protein expression levels of TGF-β1-induced FGF-2 gene were significantly down-regulated(mRNA and protein were down-regulated by 4.30 times and 1.77 times respectively).Conclusion By inhibiting fibroblast growth factor 2,matrine prevents fibroblast from accumulating in peritoneal mem-brane,and finally prevents the occurrence of peritoneal fibrosis.
