摘 要:目的分析三叶青黄酮调节PI3K/Akt-eNOS信号通路对膀胱癌细胞生物学行为的影响及其机制。方法将对数生长期T24细胞分为空白对照组、高浓度组、低浓度组,空白组在处理时采用空白培养基培养,高浓度组和低浓度组细胞在处理时采用100μg/mL和50μg/mL浓度三叶青黄酮干预;检测细胞增殖、凋亡、迁移和侵袭活性,检测细胞中PI3K、Akt、p-Akt、eNOS、β-actin蛋白表达水平。结果空白组、低浓度组和高浓度组T24细胞增殖、凋亡、迁移和侵袭活性存明显差异,其中高浓度组细胞增殖、迁移和侵袭活性最低,高浓度组细胞凋亡活性最高,且组间两两比较差异有统计学意义(P<0.05);Hoechest染色结果显示,空白组细胞细胞核相对较为完整,且颜色均一,低浓度组和高浓度组细胞细胞核出现坍缩,出现裂解碎块并可见凋亡小体,且随给药浓度增高,细胞凋亡越高;空白组、低浓度组和高浓度组T24细胞PI3K、p-Akt、eNOS水平存明显差异,其中高浓度组细胞PI3K、pAkt、eNOS水平最低,且组间两两比较均存统计学意义(P<0.05)。结论三叶青黄酮可通过调节PI3K/Akt-eNOS信号通路实现抑制膀胱癌细胞增殖、迁移和侵袭,促进细胞凋亡。Objective To analyze the effects of cloverleaf flavonoids on the biological behavior of bladder cancer cells by regulating PI3K/Akt-eNOS signaling pathway and its mechanism.Methods The T24 cells in logarithmic growth phase were divided into the blank control group,the high concentration group and the low concentration group.The blank control group was cultured in the blank medium during treatment,and the cells in the high concentration group and the low concentration group were treated with 100μg/mL and 50μg/mL of cloverleaf flavonoids.The proliferation,apoptosis,migration and invasion activities of cells were detected,and the protein expression levels of PI3K,Akt,p-Akt,eNOS andβ-actin were detected.Results Significant differences in the proliferation,apoptosis,migration and in-vasion activities of T24 cells were observed in the blank group,the low concentration group and the high concentration group,among which the proliferation,migration and invasion activities of T24 cells in the high concentration group were the lowest,and the apoptosis activity in the high concentration group was the highest,with statistical significance in pair comparisons between groups(P<0.05).Hoechest staining results showed that the cell nuclei in the blank control group were relatively complete and uniform in color.Cell nuclei in the low concentration group and the high concentra-tion group collapsed,lysed fragments and apoptotic bodies were visible,and the apoptosis of cells was increased with the increase of the drug concentration.Significant differences in the levels of PI3K,p-Akt and eNOS in T24 cells were observed of the blank control group,the low concentration group and the high concentration group,among which the levels of PI3K,p-Akt and eNOS in the high concentration group were the lowest,with statistical significance in pair comparisons between groups(P<0.05).Conclusion Cloverleaf flavonoids can inhibit the proliferation,migration and in-vasion of bladder cancer cells by regulating PI3K/Akt-eNOS signaling pathway,and promo
