摘 要:建立高效稳定的紫果西番莲植株再生体系,可为其离体快繁、倍性育种及基因工程等提供前期基础,因此以紫果西番莲的子叶为材料,基于组织培养以直接和间接的方式诱导紫果西番莲器官发生,并对得到的不定芽进行增殖、生根、炼苗移栽。结果显示:由子叶诱导得到的愈伤组织在1 mg/L 2,4-D+1 mg/L TDZ的MS培养基中通过间接器官发生途径诱导出不定芽,分化率为66.7%;子叶在1.0 mg/L 6-BA+0.5 mg/L IBA的MS培养基中能通过直接器官发生途径分化出不定芽,分化率为83.3%;最适合不定芽生根的配方为1/2 MS+0.5 mg/L NAA+0.1 mg/L IBA+0.2 g/L碳粉,生根率达83.3%;将生根苗于室内炼苗15 d后移栽,其中以红土︰腐殖土︰珍珠岩=1︰2︰1的基质配比效果最好,成活率为93.3%。根据研究结果建立紫果西番莲不定芽发生的2种途径,进一步完善了紫果西番莲的植株再生体系,可为紫果西番莲的优质种苗快繁、种质创新、遗传转化等相关研究提供技术支撑。The establishment of an efficient and stable plant regeneration system of Passiflora edulis can provide a preliminary basis for in vitro rapid propagation,ploidy breeding and genetic engineering.In this paper,the cotyledons of P.edulis were used as materials,the organogenesis of P.edulis was induced directly and indirectly based on tissue culture,and the adventitious buds were multiplied,rooting,seedling refining and transplanting.The results showed that the callus differentiation rate of cotyledons was the highest in MS medium supplemented with 1 mg/L 2,4-D and 1 mg/L TDZ,which was 66.7%.Cotyledons directly differentiated adventitious buds on MS medium supplemented with 1.0 mg/L 6-BA and 0.5 mg/L IBA,and the differentiation rate was 83.3%.The rooting rate of test-tube plantlets reached 83.3%acid in a 1/2 MS medium with 0.5 mg/L NAA 0.1 mg/L IBA and 0.2 g/L carbon powder.The rooting seedlings were transplanted after 15 days,and grew best in the soil of red soil︰humus soil︰perlite=1︰2︰1,and the survival rate was 93.3%.In this paper,two ways of adventitious bud formation of P.edulis were established,and the plant regeneration system of P.edulis was further improved,which provided technical support for high-quality seedling rapid propagation,germplasm innovation,genetic transformation and so on.
