摘 要:为减轻德宏州油茶炭疽病的危害和保证油茶产品质量,特采集该地区健康叶片,用组织分离法分离内生细菌,并通过平板对峙培养法测定所分离内生菌对油茶炭疽菌的抑制活性,后回接离体叶片对具有显著抑菌效果的内生菌验证其防效,最后用形态学及16S rRNA序列分析对抑菌效果最好的菌株进行鉴定,并通过PCR扩增脂肽基因。结果显示,从健康油茶叶片中共分离到25株内生细菌,其中菌株6715对油茶炭疽菌平板抑制效果最好,抑制率为57.59%,回接叶片发现其本身不致病,并显著抑制炭疽菌发生,经形态学和16S rRNA序列分析鉴定为Bacillus tequilensis,且扩增出脂肽基因bacA、ppsD和srfAA,具有潜在的生防价值。In order to reduce the anthracnose disease damage of Camellia oleifera and ensure the quality of C.oleifera products in Dehong Prefecture in Yunnan Province,healthy leaves of C.oleifera collected,and endophytic bacteria were isolated by tissue separation method,then the antagonistic activity of isolated endophytic bacterial strains against Colletotrichum fructicola was performed using the dual-culture plate approach,subsequently,morphological and 16 S rRNA sequence analysis were used to identify the strains with the best bacteriostatic effect,and its lipopetide antibiotic biosynthesis genes were analyzed.The results showed that 25 bacteria strains were isolated from healthy leaves of C.oleifera.Successive screenings of these 25 isolates yielded strain 6715 as the best antagonist against Colletotrichum fructicola with an efficacy of 57.59%in vitro and significantly reduced the anthracnose disease in C.oleifera detached leaves.Then the strain 6715 was identified as Bacillus tequilensis.Furthermore,PCR detection showed that the strain 6715 had the lipopetide antibiotic biosynthesis genes:bacA,ppsD and srfAA.Using the B.tequilensis strain 6715 may have the potential biocontrol value for prevention C.oleifera anthracnose disease.
