摘　　要：目的建立高效液相色谱(HPLC)法同时测定清开灵颗粒中绿原酸、栀子苷、黄芩苷、(R,S)-告依春、胆酸、猪去氧胆酸含量的方法。方法采用Thermo Syncronis-C18色谱柱(4.6mm×250mm,5 m),以乙腈-缓冲盐体系(0.05 M磷酸二氢钾水溶液,加磷酸调节pH(2.0±0.05))为流动相,梯度洗脱,流速为1.0 mL/min,采用变换波长法。结果绿原酸、栀子苷、黄芩苷、(R,S)-告依春、胆酸、猪去氧胆酸分别在0.4454~22.2720 g/mL、0.8192~40.9600 g/mL、4.4356~221.7820 g/mL、0.0803~4.0160 g/mL、4.5680~228.4000 g/mL、4.4602~223.0116 g/mL范围内与峰面积线性关系良好,平均加样回收率为99.00%,RSD=0.62%。结论 HPLC法准确度高,重复性好,可用于清开灵颗粒的的质量控制。Objective To establish a high performance liquid chromatography(HPLC)method for determining chlorogenic acid,geniposide,baicalin,(R,S)-goitrin,cholalic acid and hyodeoxycholic acid in Qingkailing Granules. Methods HPLC experiments were performed on Thermo Syncronis-C18 column(4.6 mm× 250 mm,5 m),the mobile phase was composed of acetonitrile and 0.05 M potassium dihydrogen phosphate solution which was adjusted to p H(2.0±0.05)with phosphoric acid with gradient elution at a flow rate of 1.0 m L/min. Conversion wavelength method was adopted. Results The good linearity with peak ares was achieved in the range of 0.4454~22.2720 g/m L for chlorogenic acid,0.8192~40.9600 g/m L for geniposide,4.4356~221.7820 g/m L for baicalin,0.0803~4.0160 g/m L for(R,S)-goitrin,4.5680~228.4000 g/m L for cholalic acid and4.4602~223.0116 g/m L for hyodeoxycholic acid,the average recovery was 99.00%,RSD=0.62%. Conclusion HPLC is accurate and reproducible,which can be used for the overall assessment of the quality of Qingkailing Granules.
