消瘀跌打药酒制剂和其主药通城虎药材中马兜铃酸Ⅰ的成分分析

摘  要:目的:建立消瘀跌打药酒制剂和其主药通城虎药材中马兜铃酸Ⅰ的含量测定方法。方法:采用高效液相色谱法。选用Inertsil ODS-XP(4.6 mm×150 mm,5μm)色谱柱,甲醇-0.05%冰醋酸溶液(53∶47)为流动相洗脱,检测波长:250 nm,流速:1 mL·min^(-1),柱温:30℃。结果:马兜铃酸Ⅰ的进样浓度在3.884~38.84μg·mL^(-1)范围内与峰面积积分值呈良好的线性关系,回归方程Y=55825X-12839(r=0.9999);消瘀跌打药酒中马兜铃酸Ⅰ的回收率平均为99.08%,RSD为0.61%;通城虎中马兜铃酸Ⅰ的回收率平均为98.65%,RSD为1.58%。结论:建立的消瘀跌打药酒制剂和通城虎药材中马兜铃酸Ⅰ含量测定方法,简便,重现性良好,可用于药材与制剂的质量控制。Objcetivet To establish the method for the content determination of aristolochic acid Ⅰ in Xiaoyudieda medicinal liquor and Aristolochia fordiana Hemsl.Methods HPLC method.Chromatographic conditions were as follows:Inertsil ODS-XP(4.6 mm×150 mm,5μm)column was used with mobile phase consisted of methanol-0.05%glacial acetic acid(53∶47)at a flow rate of 1.0 mL·min^(-1)under a detection wavelength at 250 nm.The column temperature was 30℃.Results The linear range of aristolochic acid Ⅰ was 3.884~38.84μg·mL^(-1),and the regression equation was Y=55825X-12839(r=0.9999).The average recovery rate of aristolochic acid Ⅰ in Xiaoyudieda medicinal liquor was 99.08%,RSD was 0.61%.The average recovery rate of aristolochic acid Ⅰ in Aristolochia fordiana Hemsl was 98.65%,RSD was 1.58%.Conclusion The method established in the experiment can be used to determine the content of aristolochic acid Ⅰ in Xiaoyudieda medicinal liquor and Aristolochia fordiana Hemsl.There were significant differences in aristolochic acid Ⅰ content between different producing areas and different batches.
