摘 要:为探究新城疫病毒(Newcastle disease virus,NDV)的V蛋白拮抗宿主干扰素(interferon,IFN)信号通路对先天性免疫的影响,本试验测定了低毒力LaSota株插入BC株V蛋白的重组病毒rL-BC的毒力和感染后4,8,12h的感染效率,病毒感染后上清的IFN-β生成情况和P-STAT1、MDA5蛋白的表达水平,并检测感染后DF1、A549细胞先天性免疫分子的转录水平。结果显示,重组病毒的颅内致病指数(intracerebral pathogenicity index,ICPI)升高,不同时间的感染效率均高于亲本毒。此外,V蛋白的插入使上清中IFN-β生成量降低,P-STAT1和MDA5蛋白的表达水平下降,LGP2、TLR3等多种模式识别受体及ISG15、IFN-α等IFN通路相关分子的转录水平下降。以上均表明NDV的V蛋白通过抑制先天性免疫反应从而提高病毒毒力。The V protein of Newcastle disease virus(NDV)can antagonize the innate immune response mediated by the interferon(IFN)signaling pathway of a host.In order to explore the effect of V protein on host innate immunity,this study determined the virulence of the recombinant virus rL-BC constructed by inserting the V protein of BC strain into the lentogenic LaSota strain and the efficiency of infection at 4,8,12hafter infection.The production of IFN-β,P-STAT1,and MDA5in DF1cells and A549cells after infection were detected.Finally,the transcriptional level of innate immune molecules in DF1,A549cells after infection was detected.The results showed that the intracranial pathogenicity index(ICPI)of the recombinant virus increased,and the infection efficiency of the recombinant virus rL-BC was higher than that of its parent virus at different times.In addition,the insertion of V protein could down-regulate the expression of IFN-β,P-SATA1,and MDA5.The results also showed that the transcriptional level of pattern recognition receptors such as LGP2and TLR3was down-regulated,and the transcriptional level of the interferon pathway-related molecules such as ISG15and IFN-αwas down-regulated.In conclusion,the V protein of NDV can increase the virulence of rL-BC by inhibiting the innate immune response.
