摘　　要：5-羟色胺(5-hydroxytryptamine,5-HT)是一种神经递质,广泛分布于哺乳动物的雌性生殖组织中,在细胞增殖、分化、卵母细胞成熟和胚胎发育等过程中起着至关重要的作用,但5-HT对猪胚胎发育的影响尚不清楚。本试验以猪孤雌激活胚胎为研究对象,利用荧光定量PCR、免疫荧光染色等技术,探讨5-HT对胚胎发育的影响。结果表明,5-HT处理可显著抑制猪孤雌胚胎的卵裂率及囊胚率,增加囊胚细胞凋亡数量,促进促凋亡基因BAX的表达,并影响H3K4me2及H3K4me3的表达。此外,5-HT处理可导致猪孤雌胚胎多能性基因NANOG显著下调,而SOX2显著上调。综上所述,5-HT在表观遗传水平可改变组蛋白甲基化,进而调控基因表达来影响猪孤雌胚胎发育。5-hydroxytryptamine(5-HT)is a neurotransmitter which is widely distributed in the female reproductive tissues of mammals,it plays a vital role in the development of cell proliferation,differentiation,oocyte maturation and embryonic development.However,the effect of 5-HT on porcine embryo development is still unclear.In this study,the effects of 5-HT on parthenogenetic activated embryos in pigs were explored by fluorescence quantitative PCR,immunofluorescence staining assay and so on.The results showed that 5-HT treatment significantly inhibited the cleavage rate and blastocyst rate of porcine parthenogenetic embryos,increased the number of blastocyst apoptosis,promoted the expression of pro-apoptotic gene BAX,and affected H3K4me2and H3K4me3levels.In addition,5-HT treatment resulted in a significant downregulation of pluripotency gene NANOG,while the expression of SOX2significantly upregulated.In summary,5-HT can affect porcine parthenogenetic embryonic development by affecting histone methylation and regulating gene expression.
