不同剂量消痔灵注射于大鼠肌肉组织中的实验研究

摘　　要：目的观察不同剂量消痔灵注射液注入大鼠肌肉组织后的反应。方法将96只大鼠随机分成正常组、1 mL注射组、2.5 mL注射组与5 mL注射组,将相应量消痔灵注射液原液注入鼠左后肢内收肌处,在注射后第3天、第7天、第14天与第21天,取注射部位肌肉组织作苏木精-伊红(HE)染色,观察肌肉组织变化过程。结果各组大鼠不同时间段观察均反应敏捷,呼吸、饮食、体重均正常,均未见死亡。1 mL注射组与正常组,左后肢均未见肿胀、活动正常。HE染色除1 mL注射组第21天筋膜见炎症组织浸润,余均未见明显异常;2.5 mL注射组于不同时间段逐渐出现左后肢内收肌处肿胀伴足背肿胀,活动不受限,HE染色见肌间隙逐渐增宽,炎症逐渐向周围筋膜浸润;5 mL注射组不同时间段左后肢内收肌处肿胀伴足背肿胀均较2.5 mL注射组明显,活动逐渐受限,HE染色见肌组织肿胀,炎症不断明显并逐渐向周围筋膜浸润,至第21天见肌组织固缩,肌间隙增宽,周边组织坏死。结论大剂量消痔灵注射液原液注射于肌肉组织中,易引起组织坏死,注射消痔灵时须注意。【Objective】To observe the response of injecting different doses of Xiaozhiling into muscle tissue of rats.【Methods】Ninety-six rats were randomly divided into normal group,1 mL injection group,2.5 mL injection group and 5 mL injection group.The corresponding dose of Xiaozhiling was injected into adductor muscle of left hind limb of rats.On the 3 rd,7 th,14 th and 21 st day after injection,the muscle tissues of injection site were taken for HE staining,and the changes of these muscle tissues were observed.【Results】The rats in each group were quick,breathing,eating and weight were normal,no death was observed in different periods.There was no swelling in the left hind limb of the rats and normal movement was observed in the 1 mL injection group and the normal group.Except for the infiltration of inflammatory tissue of fascia in 1 mL injection group on the 21 st day,no abnormality was found.At different time,the adductor muscle of the left hindlimb of the rats in the 2.5 mL injection group gradually appeared swelling with swelling of the dorsum of the foot,with unrestricted movement.The HE staining showed that the muscle space gradually widened,and the inflammation gradually infiltrated into the surrounding fascia.In different time periods,the adductor muscle of left hindlimb in 5 ml injection group gradually swelled with the swelling of the back of foot,which was more obvious than that in 2.5 mL injection group.The activity was gradually limited.The HE staining showed that the muscle tissue swelled,the inflammation was continuously obvious and gradually infiltrated into the peripheral fascia.On the 21 st day,the muscle tissue was pyknosis,the muscle space was widened,and the peripheral tissue was necrotic.【Conclusion】Injection of large dose of Xiaozhiling into muscle tissue is easy to cause tissue necrosis,which should be paid attention to in clinical injection.
