摘 要:目的:观察自拟清热凉血方对牙周炎大鼠的抑炎作用及对牙周组织生长的影响,并探讨Toll样受体4(TLR4)通路在其中发挥的作用。方法:85只大鼠随机取70只建立牙周炎大鼠模型,建模成功64只大鼠随机分为模型对照组、低剂量组、高剂量组和抑制剂组,每组16只,剩余15只为正常对照组。观察各组大鼠一般情况及牙龈指数;酶联免疫法检测肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)水平;HE染色观察牙龈组织病理学;RT-qPCR检测TLR4 mRNA、髓样分化因子88(MYD88)mRNA、核因子κB p65(NF-κB p65)mRNA相对表达量;Western blotting检测TLR4、MYD88、NF-κB p65和p-NF-κB p65蛋白相对表达量。结果:正常对照组大鼠牙龈指数为0;与模型对照组比较,低剂量组、高剂量组和抑制剂组大鼠牙龈指数明显降低,且抑制剂组<高剂量组<低剂量组(P<0.01)。与正常对照组比较,模型对照组、低剂量组、高剂量组和抑制剂组大鼠血清TNF-α、IL-1β水平升高,且抑制剂组<高剂量组<低剂量组<模型对照组(P<0.01或P<0.05)。与模型对照组比较,低剂量组、高剂量组和抑制剂组大鼠一般情况、牙周组织异常病理变化减轻,其中抑制剂组改善更显著。与正常对照组比较,模型对照组、低剂量组、高剂量组和抑制剂组大鼠牙周组织TLR4 mRNA、MYD88 mRNA及蛋白相对表达量升高,p-NF-κB p65蛋白相对表达量升高,且抑制剂组<高剂量组<低剂量组<模型对照组(P<0.05)。结论:自拟清热凉血方可减轻牙周炎大鼠炎症反应,促进牙周组织生长,机制可能与抑制TLR4通路有关。Objective:To observe the anti-inflammatory effect of Zini Qingre Liangxue Prescription on periodontitis rats and its influence on periodontal tissue growth,and to explore the role of Toll-like receptor 4(TLR4)pathway in it.Methods:A total of 70 rats were randomly selected from 85 rats to establish periodontitis model.64 rats were successfully modeled and randomly divided into model control group,low-dose group,highdose group,and inhibitor group,with 16 rats in each group.The remaining 15 rats were the normal control group.The general condition and gingival index were observed.Enzyme-linked immunoassay to detect tumor necrosis factor(TNF-α)and interleukin 1β(IL-1β)levels.The gingival histopathology was observed by HE staining.RT-qPCR to detect the relative expression of TLR4 mRNA,myeloid differentiation factor 88(MYD88)mRNA,and nuclear factor kappa B p65(NF-κB p65)mRNA.The relative expression of TLR4,MYD88,NF-κB p65 and p-NF-κB p65 protein was detected by Western blotting.Results:The gingival index of rats in the normal control group was 0;compared with the model control group,the gingival index of rats in the low-dose group,high-dose group and inhibitor group was significantly reduced,and the inhibitor group<high-dose group<low-dose group(P<0.01).Compared with the normal control group,the serum TNF-α and IL-1β levels of the model control group,low-dose group,high-dose group and inhibitor group increased,and the inhibitor group<high-dose group<low-dose group<model control group(P<0.01 or P<0.05).Compared with the model control group,the general condition and abnormal pathological changes of the periodontal tissue of the rats in the low-dose group,high-dose group and inhibitor group were reduced,and the inhibitor group improved more significantly.Compared with the normal control group,the mRNA and protein expression of TLR4 and MyD88 in model control group,low-dose group,high-dose group and inhibitor group increased,and the protein expression of p-NF-κB p65 increased,and inhibitor group<high-dose group<low
