摘  要:目的探讨小儿活血止痛颗粒对急性软组织损伤的药效作用。方法将SPF级昆明种小鼠随机分为空白对照组(生理盐水)、阳性对照组(0.25 g/kg云南白药),以及小儿活血止痛颗粒低、中、高剂量组(分别相当于原药剂量2.31,4.62,6.94 g/kg),各10只。各组小鼠每天灌胃1次相应药物,连续给药7 d,采用醋酸致小鼠扭体模型和热刺激法致小鼠疼痛模型,考察镇痛作用;采用二甲苯致小鼠耳廓肿胀模型,考察消肿作用。取SPF级健康雄性SD大鼠,采用机械冲击挫伤法制备急性软组织损伤模型。将100只建模成功的大鼠随机分为模型组(B组,生理盐水),阳性对照组(C组,0.2 g/kg云南白药),以及小儿活血止痛颗粒高、中、低剂量组(D_(1)组、D_(2)组、D_(3)组,相当于原药剂量7.00,3.50,1.75 g/kg),各20只;同时选择正常大鼠20只作为正常组(A组,生理盐水)。各组大鼠每天灌胃1次相应药物,连续给药7 d,观察其组织损伤程度,并测定血液流变学参数。结果与空白对照组比较,小儿活血止痛颗粒中、高剂量组醋酸所致疼痛小鼠的扭体潜伏期均显著延长,扭体次数均显著减少(P<0.05);小儿活血止痛颗粒高剂量组热刺激所致疼痛小鼠末次给药1.5 h后痛阈时间显著延长(P<0.05);小儿活血止痛颗粒中、高剂量组二甲苯所致耳廓肿胀小鼠的耳廓肿胀度均显著降低(P<0.05)。对于机械冲击挫伤致急性软组织损伤,与B组大鼠比较,D_(1)组、D_(2)组、D_(3)组大鼠的损伤证候评分分别在给药第3,5,7天显著降低(P<0.05);D_(1)组、D_(2)组大鼠的全血黏度和血浆黏度均在给药第3天显著降低(P<0.05)。结论小儿活血止痛颗粒具有消肿、镇痛、活血、化瘀功效,对软组织损伤有显著改善作用。其消肿、镇痛作用可能与减轻炎性因子渗出和肿胀有关,活血化瘀作用可能与降低全血黏度和血浆黏度有关。Objective To investigate the pharmacodynamics of the Xiaoer Huoxue Zhitong Granules on acute soft tissue injury.Methods SPF Kunming mice were randomly divided into blank control group(normal saline),positive control group(0.25 g/kg Yunnan Baiyao)and Xiaoer Huoxue Zhitong Granules low-,medium-and high-dose groups(equivalent to 2.31,4.62 and 6.94 g/kg of crude drug,respectively),10 mice in each group.The mice in each group were given corresponding drugs by intragastric administration once a day for 7 d.The analgesic effect of Xiaoer Huoxue Zhitong Granules was evaluated by using acetic acid-induced writhing model and thermal stimulation-induced pain model.The swelling-subsiding effect of Xiaoer Huoxue Zhitong Granules was evaluated by using dimethylbenzene-induced auricle swelling model.SPF healthy male SD rats were selected to establish the model of acute soft tissue injury by mechanical shock contusion method.A total of 100 rats with successful modeling were randomly divided into normal group model group(group B,normal saline),positive control group(group C,0.2 g/kg,Yunnan Baiyao),and Xiaoer Huoxue Zhitong Granules high-,medium-and low-dose groups(groups D_(1),D_(2) and D_(3),equivalent to 7.00,3.50 and 1.75 g/kg of crude drug),20 rats in each group.At the same time,20 normal rats were selected as normal group(group A,normal saline).The rats in each group were given corresponding drugs by intragastric administration once a day for 7 d.The degree of tissue injury was observed,and the hemorheological parameters were measured.Results Compared with those in the blank control group,the writhing latency of mice with acetic acid-induced pain significantly prolonged,and the writhing frequency significantly reduced in the Xiaoer Huoxue Zhitong Granules medium-and high-dose groups(P<0.05).Compared with that in the blank control group,the pain threshold time of mice with thermal stimulation-induced pain in high-dose Xiaoer Huoxue Zhitong Granules groups significantly prolonged at 1.5 h after the last administration(P<0.05).
