前列腺癌组织中miR-449a、miR-493-5p表达与临床病理参数及预后的关系

摘　　要：目的检测miR-449a、miR-493-5p在前列腺癌组织中的表达水平,探究miR-449a、miR-493-5p表达与前列腺癌患者临床病理参数和预后之间的关系。方法选取2013年2月至2015年3月在常州市第七人民医院住院的125例前列腺癌患者的石蜡组织样本及其相应的癌旁组织样本作为研究对象。采用实时荧光定量PCR(RT-qPCR)实验检测组织中miR-449a、miR-493-5p表达水平,同时分析miR-449a和miR-493-5p表达与患者临床病理参数之间的关系,分析miR-449a、miR-493-5p表达与患者预后关系。结果与癌旁正常组织相比,前列腺癌组织中miR-449a、miR-493-5p表达降低,差异具有统计学意义(P<0.05)。前列腺癌组织中miR-449a表达水平与Gleason评分、骨转移及肿瘤TNM分期有关(P<0.05);miR-493-5p表达水平与前列腺特异性抗原(PSA)、Gleason评分、骨转移及肿瘤TNM分期有关(P<0.05)。生存分析结果显示,miR-449a高表达患者的术后5年总生存率高于miR-449a低表达患者,差异具有统计学意义(P<0.05);miR-493-5p高表达患者的5年总生存率高于miR-493-5p低表达患者,差异具有统计学意义(P<0.05)。结论前列腺癌组织中miR-449a和miR-493-5p均表现为低表达,且与部分临床病理参数相关,两者对前列腺癌患者的预后均有一定的预测价值。Objective To detect the expression of miR-449 a and miR-493-5 p in tissue samples of patients with prostate cancer, and to study the relationship between the two and clinicopathological parameters and prognosis. Methods Paraffin tissue samples and their adjacent tissues were collected from 125 patients with prostate cancer who were admitted to Changzhou Seventh People′s Hospital from February 2013 to March 2015. The expression levels of miR-449 a and miR-493-5 p in tissue samples were detected by real-time fluorescent quantitative PCR(RT-qPCR) technology. The relationship between miR-449 a, miR-493-5 p expression and clinical characteristics of patients was analyzed, and the relationship between the expression of miR-449 a, miR-493-5 p and the prognosis of prostate cancer patients was analyzed. Results Compared with adjacent normal tissues, the expression levels of miR-449 a and miR-493-5 p in prostate cancer tissues were decreased(P<0.05). The expression level of miR-449 a in prostate cancer tissues was related to Gleason score, bone metastasis and tumor TNM stage(P<0.05), the expression level of miR-493-5 p was related to prostate-specific antigen(PSA), Gleason score, bone metastasis and tumor TNM stage(P<0.05). The survival analysis results showed that the 5-year survival rate of patients with high expression of miR-449 a was higher than those of patients with low expression of miR-449 a(P<0.05), and the 5-year overall survival rate of patients with high expression of miR-493-5 p was higher than that of patients with low expression of miR-493-5 p(P<0.05). Conclusions The expressions of miR-449 a and miR-493-5 p in prostate cancer are low, which are related to some clinicopathological parameters. Both of them have certain predictive value for the prognosis of patients.
