摘 要:目的探讨七氟醚促进宫颈癌细胞迁徙转移的机制,并在此基础上探索治疗药物。方法选取2018年5月购自ATCC公司的人宫颈癌细胞Hela细胞作为研究对象。利用七氟醚诱导Hela细胞作为模型组,七氟醚及JAK激酶抑制剂乌帕替尼共培养作为药物组。分别利用Transwell、Westernblot及ELISA试剂盒检测空白组、模型组及药物组细胞的迁徙转移能力、P-STAT3及P-JAK蛋白表达及细胞上清中IL-6、IL-1β及TNF-α的表达。结果与空白组比较,模型组细胞迁移能力显著增强,细胞上清中IL-6、TNF-α浓度及细胞内STAT3、JAK蛋白磷酸化水平的表达均显著升高,差异具有统计学意义(P<0.05);JAK激酶抑制剂显著抑制了七氟醚诱导的Hela细胞迁徙转移,同时明显降低了细胞内JAK及STAT3蛋白磷酸化水平及上清中IL-6的浓度。结论JAK激酶抑制剂可以通过作用于IL-6/JAK/STAT3通路改善由七氟醚诱导的宫颈癌转移。Objective To mechanism of sevoflurane induced migration of Hela cells and screen the therapeutic drugs. Methods Human cervical cancer cells Hela cells purchased from ATCC in May 2018 were selected as the research object. Hela cells were induced by sevoflurane as the model group, and co-culture of sevoflurane and a JAK inhibitor upadacitinib was used as the drug group. Transwell, Western Blot and ELISA kit were used to detect the migration and metastasis ability of the blank group, model group and drug group, the protein expression of p-STAT3 and p-JAK, and the expression of IL-6, IL-1β and TNF-α in the supernatant of the cells. Results Compared with blank group, the cell migration ability of model group was significantly enhanced, the concentrations of IL-6 and TNF-α in cell supernatant and the expression of intracellular STAT3 and JAK protein phosphorylation were significantly increased, with statistical significance(P<0.05). JAK inhibitor significantly inhibited sevoflurane-induced Hela cell migration and decreased the phosphorylation levels of JAK and STAT3 proteins and the concentration of IL-6 in the supernatant. Conclusions JAK inhibitors can inhibit sevoflurane-induced cervical cancer metastasis by acting on the IL-6/JAK/STAT3 pathway.
