玉米细菌性枯萎病菌检测引物/探针的特异性评估

摘　　要：利用6株玉米细菌性枯萎病菌(Pantoea stewartii subsp.stewartii,PSS)和29株相关菌株对报道的18对检测引物/探针的特异性进行测试。试验结果显示,其中16对引物/探针会出现假阳性或弱假阳性扩增,仅引物cpsAB2313F/cpsR和DC283galE/DC283galEc可以特异检测目的菌,两对引物的检测灵敏度分别为0.1 pg和1 pg。根据测试结果,推荐这2对引物用于检测玉米细菌性枯萎病菌,优先推荐引物cpsAB2313F/cpsR。6 strains of Pantoea stewartii subsp.stewartii and 29 strains of other related strains were collected,and 18 pairs of specific PCR primers/probes for the detection of P.stewartii subsp.stewartii were tested.The results showed that 16 pairs primers/probes had false positive amplification or weak false positive amplification.Only the cpsAB2313 F/cpsR and DC283 galE/DC283 galEc primers had good specificity with the detection sensitivity of 0.1 pg and 1 pg.Based on the above results,the cpsAB2313 F/cpsR and DC283 galE/DC283 galEc primers are recommended for the detection of P.stewartii subsp.stewartii,and the cpsAB2313 F/cpsR primers is better than the other.
