荷兰进境毛蕊银莲花中毁芽滑刃线虫的形态和分子鉴定

摘　　要：宁波口岸从荷兰进境的毛蕊银莲花种苗中发现一种滑刃属线虫活虫,该群体的主要形态特征为:侧线4条,雌虫尾近圆柱形,末端有尾尖突,长1.6-2.2μm,其上有多个微小瘤状突,后阴子宫囊显著,长77.7-121.3μm。雄虫尾端有1.7-2.5μm的尾尖突,交合刺为玫瑰刺状,有显著的基顶和喙突,背缘远端腹弯,形成小勾状,中弧线长19.6-25.7μm。基于部分18S和28S基因DNA序列构建的系统发育树显示,该群体与已知毁芽滑刃线虫亲缘关系近且在同一进化分支中。因此,将该种线虫鉴定为毁芽滑刃线虫(Aphelenchoides blastophthorus Franklin,1952)。这是我国口岸首次截获该线虫。A number of living Aphelenchoides were isolated in Anemone cathayensis Kitag.var.hispida Tamura from Netherland at Ningbo port.The morphological characteristics of the population were mainly as follows:presence of four lines in the lateral field;a sub-cylindrical female tail bearing a ventrally located single mucron,1.6-2.2μm long,with a number of tiny strumaes on it;well-developed postuterine sac,77.7-121.3μm long.Similar to females,the males tail carries a 1.7-2.5μm simple mucron.The spicules curved ventrally and rose thorn shaped,condyles and rostrum well developed,dorsal limp ventrally curved and forms a small hook at distal end,spicule length(in curved median line)19.6-25.7μm.The neighbour-joining trees of DNA sequences from partial 18 S and 28 S region demonstrated that the population and A.blastophthorus had very close relationship and located in the same branch.Based on the morphological characteristics and molecular analysis,the population from Netherland was identified as Aphelenchoides blastophthorus Franklin,1952.This is the first interception report in China.
