摘 要:目的:观察芪术抗纤颗粒对肺纤维化大鼠肺组织病变的防治作用,并初步分析其作用机制。方法:建立博来霉素诱导的大鼠肺纤维化模型,早期干预组从第1天、治疗组从第14天予芪术抗纤颗粒灌胃。观察第3、7、14、28天大鼠肺组织的炎症和纤维化程度、α-平滑肌肌动蛋白(α-SMA)表达水平,肺泡灌洗液中肿瘤坏死因子α(TNF-α)和转化生长因子β1(TGF-β1)的水平。结果:与模型组同期比较,早期干预组第14、28天肺泡炎,肺组织α-SMA表达水平,肺泡灌洗液TGF-β1水平及第14天肺泡灌洗液TNF-α水平均显著降低(P<0.05)。治疗组治疗后与本组治疗前比较,肺泡炎和胶原纤维增生程度,肺组织α-SMA表达水平,肺泡灌洗液TNF-α水平均显著下降(P<0.05)。结论:芪术抗纤颗粒对肺纤维化大鼠肺组织炎症反应和肺纤维化病变具有一定的防治作用,其机制可能与降低肺组织炎性细胞因子和促纤维化细胞因子水平有关。Objective:To observe the preventive and therapeutic effect of Qizhu Kangxian Granules on pulmonary tissue of rats with pulmonary fibrosis,and to analyze its mechanism preliminarily.Methods:Bleomycin-induced pulmonary fibrosis model in rats was used.Rats of early intervention group were given Qizhu Kangxian Granules gavage from the first day when rats of treatment group were given from the 14th day.We observed the degree of inflammation,fibrosis,the expression level ofα-SMA,and the levels of TNF-αand TGF-β1 in BALF of rats on the 3rd,7th,14th and 28th days.Results:Compared with model group at the same time,the degree of alveolitis,the expression level ofα-SMA in lung tissue,the level of TGF-β1 in BALF at the 14th and 28th day,and the level of TNF-αin BALF at the 14th ady in the early intervention group decreased significantly(P<0.05).After treatment,the degree of alveolitis,collagen hyperplasia,the expression level ofα-SMA in lung tissue and the TNF-αlevel in BALF in treatment group decreased significantly(P<0.05).Conclusion:Qizhu Kangxian Granules has a certain prevention and treatment effect on the inflammatory response of lung tissue and the pathological changes of lung fibrosis in rats.Its mechanism may be related to the decrease of inflammatory and fibrogenic cytokines in lung tissue.
