摘 要:目的:观察祛风宣肺方对模型豚鼠肺组织病理及PLC-β/PKC、TRPA1/V1蛋白表达的影响。方法:将48只SPF级雄性健康Hartley豚鼠随机分为正常组、模型组、TRPV1抑制剂组、TRPA1抑制剂组、PLC抑制剂组、祛风宣肺方组,每组8只,应用经典咳嗽高敏感豚鼠模型,相应药物干预7d后,肺功能仪检测麻醉后豚鼠气道阻力,HE染色法观察肺组织病理变化,ELISA法检测BALF中SP水平,免疫组化法测定肺组织中PLC-β、PKC、TRPA1、TRPV1、NK-1R蛋白表达水平。结果:与模型组比较,各组气道阻力均显著降低(P<0.01,P<0.05),肺组织TRPA1、TRPV1、NK-1R蛋白表达显著降低(P<0.05,P<0.01),其中PLC抑制剂组与祛风宣肺方组肺组织PLC-β蛋白表达显著降低(P<0.01,P<0.05);模型组肺组织病理及BALF炎症反应明显,TRPV1抑制剂组、TRPA1抑制剂组、PLC抑制剂组及祛风宣肺方组炎症水平降低(P<0.01)。结论:祛风宣肺方可降低咳嗽敏感性增高模型豚鼠的气道反应性,减轻肺组织炎症,可能通过下调肺组织中PLC-β、TRPV1、TRPA1、NK-1R的蛋白表达水平发挥作用。Objective:To observe the effects of Qufeng Xuanfei Decoction on the pathological changes and the expression of PLC-β,PKC,TRPA1,TRPV1 proteins in lung tissue of model guinea pig.Methods:Forty-eight guinea pigs were randomly divided into blank control group,model group,TRPV1 inhibitor group,TRPA1 cnhibitor group,PLC inhibitor group,Qufeng Xuanfei group,8 in each group,the model of cough hyperactivity was established.After 7d-intervention,the airway reactivity was measured by spirometer under different concentrations of acetylcholine;the pulmonary pathology was observed by HE stain;the SP in BALF was detected by ELISA;the expression of PLC-β,PKC,TRPA1,TRPV1 proteins in lung tissue was detected by immunohistochemistry.Results:Compared with model group,the airway resistance and expression of lung TRPA1,TRPV1,NK-1R proteins in each group decreased significantly(P<0.01,P<0.05);PLC-βproteins in lung decreased significantly in PLC inhibitor group and Qufeng Xuanfei group(P<0.01,P<0.05).The lung tissue showed apparent structure destruction and airway inflammation in model group,while the TRPV1 inhibitor group,TRPA1 inhibitor group,PLC inhibitor group and Qufeng Xuanfei group showed a lower level of structure destruction and airway inflammation compared with model group(P<0.01).Conclusion:Qufeng Xuanfei Decoction could reduce the airway reactivity and inflammation of guinea pigs with cough hyperactivity,the therapeutic mechanism might be regulated by the expression of PLC-β,PKC,TRPA1,TRPV1 proteins in lung tissue.
